Objective : For providing a theoretical basis for clinical prevention and cure against AD,according to the existing pathogenesis of Alzheimer's disease to set up Alzheimer's disease model, by analyzing the praxiology,the state of oxidative stress and morphology among groups,to study the intervention effect and mechanism of integripetal rhodiola herb (IRH) against Alzheimer's disease model rats.Method:1. Model:AD model was formed by adopting the D-gal+AlCl3+SCOP on health male SD rats.2. Grouping: Reference to the literature using random number table this model success were randomly divided into three groups, namely, model group, Chinese drug(Integripetal rhodiola herb) group, western medicine (Hydergine) group, setting up a blank group.3. Delivery methods: For 28 days,Rhodiola group and Hydergine group were intragastrically given to by a dose of 5g/(kg·d) and 1mg/(kg·d) respectively. While the rats in model group and blank group were intragastically given 5ml/(kg·d) distilled water to.4. Detect indicators: Using Morris water maze to detect the ability of learning and memory of rats, including place navigation and probe trial testing;The activity of anti-O2·—and T-AOC to be tested can discover the disparity of oxidative stress among groups.5. Morphology change:Common HE staining is undertaken to observe the morphology change in the AD rats'hippocampus.Result:1.Place navigation:After the success of establishing the AD model rats,during the first day of Water maze training, the escape latency of model group was significantly longer than the other groups (P <0.05);After repeated measurements analysis of variance, there are no significant differences among blank group,IHR group ang hydergine group(P>0.05); there are significant differences between model group and other three groups;2. Probe trial testing:the percentage(the ratio between the swimming time in the quadrant including the platform and the total time) and the number of finding the platform of model group are decreasing remarkably compared with the other groups(P <0.05);there is no significant differences among the percentage of hydergine group and blank group and IRH group(P>0.05); the percentage of IRH group is smaller than the one of blank group(P<0.05); there are no significant differences of the number of finding the platform among blank group,IHR group ang hydergine group(P>0.05);3.the activity of anti-O2·—of model group is lower than blank group,IRH group and hydergine group(P<0.05); the activity of anti-O2·—of the treatment group is no remarkably different compared with blank group(P>0.05);4.the activity of T-AOC of model group is lower than the other groups(P<0.05); the activity of T-AOC of the treatment group is no remarkably different compared with blank group(P>0.05);5. Compared with the other groups,the cellula nervosa of model group rats are damaged badly,such as Granulovacuolar degeneration,decreasing of sum of cellula nervosa.Senile Plaques emerge in hippocampus,which is a kind of eosinophilic staining bolus.In addition,the neurofibril can be seen thickening,squirming and tangling.Conclusion:1. The AD model ,which was formed by injecting D-gal into abdominal cavity and taking AlCl3 by intragastric administration as well as injecting scopolamine at last,is a successful model.2.The IRH has a significantly relieving effect on the lesion against learning and memory of AD model rats,and can improve their learning and memory ability.3.The IRH has a good protecting effect on cellula nervosa of the AD rats by eliminating free radical and improving the anti-oxydation potentia.4.the change of pathology of brain tissue can be improved by IRH .
