Studise On The Effect Of Ethology,Enzymology And Apoptosis Of Alzheimer's Disease Model Rats With Yinnaoling

Objective:To verify the effect of invigorating the kidney and invigorating spleen and soothing liver and reducing phlegm by resuscitation to prevent and cure AD,and to investigate protective and therapeutic mechanism of Yinaoling on experimental AD rats.Method:AD rats model could be induced by intraperitoneral injection of D-galactose combined with amyloid-β_25-35 injection the Hippocampus.Taking the cholinesterase inhibitor as to be the checking-medicine, we studied the ability of initiative and passive learning and memory of the rats by Morris water maze and step down test;maked detection of the activity of AchE, the energy of SOD and the content of MDA by chemistry chromatometry;observed morphology by the light microscope and electron microscope;maked detection of the expression of Bcl-2 and Bax by immunochemistry tissue staining method.Eventually,the effect of Yinaoling on above-mentioned index was investigated.Result: The experimental result shows that Yinaoling has the following effects:1. ethology: compared with Blank group, delitescence of model group prolonged obviously, frequency of finding the platform decreased,number of times of mistake increased (p<0.01) .Treat group and prevent group compared with model group,delitescence shortened,frequency of finding the platform increased,number of times of mistake decreasing (p<0.01) .2. enzymology: compared with Blank group, energy of Ache of model group heightened,activity of SOD degraded, cont of MDAstepped up ( P<0.01) . Treat group and prevent group compared with model group, energy of Ache degraded,activity of SOD heightened, cont of MDA cut down (P<0.01) .3. apoptosis: compared with Blank group, area density of bcl-2 proteinum descended, area density of bax proteinum ascended(P<0.01) . Treat group and prevend group compared with model group, area density of bcl-2 proteinum ascended, area density of bax proteinum descended (P<0.01)4. morphology change: sum of neuron of model group was less than that of blank group. Cell Apoptosis phenomenon could be seen ,such as cell nucleus of some neuron showed pyknosis condition, caryotheca slightly thickened and aberration, lipofuscin increased and so on.Yinaoling treat group and prevend group had more neurons,in which the change of bioblast was not obvious,distribution of neurofibra was regular, cell organ in intracytoplasm had normal structure.Conclusion:The protective and therapeutic mechanism of Yinaoling is concerned with protecting central cholinergic system and resisting free rADical and inhibiting apoptosis.Yinaoling is an effective prescription for prevention and therapy of AD.
