The paper consists of two parts:literature reviews and experimental trials. First Section:Literature SummaryThere are three literature reviews. The first article is involved variation regularity and mechanism of 5-hydroxytryptamine (5-HT) in the brain injury, and experimental and clinical overviews about the aspects of nerve cells, blood-brain barrier; The second review discuss the mechanisms of dopamine (DA) in brain injury, and research survey of the cell toxicity; The last review highlights clinical and experimental research about acupuncture treating the brain injury.Second Section:Experimental Research Objective:To discuss the protective mechanism of electroacupuncture (EA) in the brain injury.Methods:Rats were divided into three groups randomly:Blank Group, Model Group and EA Group. The brain injury rat model was made by using a modified Feeney 's method. We will take the brain tissues at 6h,12h,24h,48h,6d after injury.(1) Comprehensive evaluation on rats neurological function after brain injury by using the Neural injury severity score (NSS) to develop the simple evaluation criteria; (2) The injured tissues of EA Group, Blank Group and Model Group were removed to detect the content of 5-HT, DA by high performance liquid chromatography and image analysis.Results:After the brain injury, the rat model was made successfully, (1) Compared with the Blank group, the NSS were increased significantly in Model Group and EA Group at 6h(P<0.01),12h(P<0.01),24h(P<0.01),48h(P<0.01), 6d(P<0.05); Compared with the Model group, the NSS were decreased significantly in EA Group at 6h(P<0.01),12h(P<0.05),24h(P<0.01),48h(P<0.01),6d(P<0.05).(2) Compared with the Model group, the 5-HT content of injury local tissues were increased significantly in EA Group and Blank Group at the corresponding times. Compared with the Blank Group, the 5-HT content of injury local tissues were increasedin EA Group at 24h,48h,6d and there was significant difference. (P<0.01); the 5-HT content of injury local tissues were increased slightly in at 6h,12h and there was no significant difference. (P>0.05). (3) Compared with the Blank group, the DA content of injury local tissues were increased significantly in Model Group (P<0.01); Compared with the Model group, the DA content of injury local tissues were decreased significantly at 6h(P<0.05), 12h(P<0.05),24h(P<0.01),48h(P<0.01),6d(P<0.05). Compared with the Blank Group, the DA content of injury local tissues were increased significantly at 12h,24h, 48h. (P<0.05); the DA content of injury local tissues were increased lightly at 6h,6d and there was no significant difference (P>0.05). Conclusion:EA has therapeutic effect on the brain injury. It maybe decrease the content of 5-HT and DA, relieve the dysfunction of synthesis and metabolism caused by the 5-HT and DA to reduce the vascular injury and neuronal toxicity. EA also has the general roles to regulate the monoamine neurotransmitters.
