Objective: To investigate whether electroacupuncture exerts its effec ts on acute ischemic brain injury by regulating changes in the glycogen synthase-3? signaling pathway.Methods: Seventy-five male SD rats were randomly divided into a total of 5 groups(n=15),which are sham operation group(Sham group),operation group(MCAO group),electroacupuncture group(EA group),GSK-3? inhibitor group(SB415286 group),and electroacupuncture+GSK-3? Inhibitor group(EA+SB group);In addition to sham group,MCAO group,EA group,SB group,EA+SB group are made middle cerebral artery occlusion model.In the sham group,the line blot block the mid dle cerebral artery which does not enter the brain.The acupuncture poi nts were selected as in the rats "Baihui and Shuigou",and the EA gro up and EA+SB group began to receive electroacupuncture stimulation f or 30 min during reperfusion.After cerebral ischemic reperfusion,the r at brain hippocampal tissue was taken off.TTC staining was used to e valuate the volume of brain infarct.TUNEL was used to detect apoptos is.Western Blot and RT-q PCR were used to detect the expression of GSK-3? protein and gene in hippocampus of each group.Resuts:(1)Changes in cerebral infarct volume: Less infarct volume was seen in the Sham group,and cerebral infarction volume was signifi cantly increased in the MCAO group after cerebral ischemia/reperfusion.The cerebral infarction volume in EA group and SB group were signif icantly lower than that in MCAO group(P<0.05).The cerebral infarctio n volume in EA+SB group was significantly decreased from that in M CAO group,SB group and EA group(P<0.05).There was no statistical ly significant difference in cerebral infarction volume between the EA g roup and the SB group(P>0.05).(2)Apoptosis changes: A small amount of apoptosis were observ ed in Sham group,and the number of apoptosis in MCAO group was significantly increased(P<0.05).The number of apoptosis in EA group and SB group were lower than that in MCAO group(P<0.05).Compar eing with EA group and SB group,the number of apoptotic cells in E A+SB group was decreased(P<0.05).There was no significant differenc e in the number of apoptotic cells between EA group and SB group(P>0.05).(3)Western Blot detection: The expression of GSK-3? protein w as slightly expressed in Sham group,and the expression of GSK-3? in hippocampus of MCAO group was significantly increased(P<0.05).The expression of GSK-3? protein in EA group and SB group were signific antly lower than that in MCAO group(P<0.05).When the GSK-3? wa s inhibited by SB415286,the electroacupuncture stimulation can effectiv ely reduce the expression of GSK-3?,and the expression of GSK-3? pr otein in EA+SB group was lower than SB group and EA group(P<0.05).There was no significant difference in GSK-3? protein expression b etween EA group and SB group(P>0.05).(4)RT-qPCR detection:The relative expression of GSK-3? mRNA was lower in Sham group,and the relative expression of GSK-3? mR NA in hippocampus of MCAO group was significantly increased(P<0.05).The relative expression of GSK-3? mRNA in EA group and SB gr oup were significantly lower than that in MCAO group(P<0.05).When the GSK-3? was inhibited by SB415286,the electroacupuncture stimulat ion can effectively reduce the relative expression of GSK-3? mRNA.Th e relative expression of GSK-3? m RNA was decreased in EA+SB grou p compared with SB group and EA group(P<0.05).There was no sign ificant difference of the relative expression of GSK-3? mRNA in EA g roup and SB group(P>0.05).Conclusions:(1)Electroacupuncture can reduce cerebral infarction vo lume and reduce apoptosis in ischemic brain injury of rat.(2)SB415286 was GSK-3 specific inhibitor,which can inhibit the activity of GSK-3 and exert its effect on in ischemic brain injury.(3)Not only electroa cupuncture exerts brain protection related to GSK-3? signaling,but also electroacupuncture exerts anti-ischemic brain damage and brain protecti on effect through inhibiting GSK-3? signaling.
