Experimental Animals Lung Qi Deficiency And Allergic Rhinitis, Immune-related Research

OBJECYIVE:To study the immune relationship between deficiency of Lung Qi and the allergic rhinitis(AR) by testing items of physiology,pathology and molecular biology through animal experiment and to provide partial experimental evidences for the clinical application of Qi-replenishing and Lung-tonifying therapy on treating AR.METHODS:Normal SD rats were divided into 4 groups:control group,model group of AR(AR group),model group of deficiency of Lung Qi and AR(Lung deficiency and AR group),model group of deficiency of Lung Qi,AR and herbal medication(Lung deficiency and AR herbal group).The model of deficiency of Lung was carried out the way of combination of Medical Experimental Animal and Practical Animal Model of TCM syndrome.Model of AR was done by report of An Yunfang.The complex model was achieved on that of deficiency of Lung Qi followed by AR model.Rats of herbal group were given medication right after abdominal injection.One hour later,rats of other groups were given 5%ovalbumin solution to be semi-activated except the control group.The general condition was observed from the second day of modeling.Nasal symptoms and secretion were focused on within 30 minutes of solution dropping.Rats were observed for 30 minutes respectively after nasal activation,to evaluate the occurrence of AR at the seventh day.Then they were executed.The maxillary bones of 8 rats were randomly chosen and stripped.The pH volume of mucosa of nasal septum was examined by the standard shade guide.The pathological morphous was viewed by light microscope and electron microscope.The serumal content of IL-2,IL-4 and IgE was determined by EIA. Additionally,the serum content of histamine of AR model rats was measured by PCA after abdominal injection to reveal the modeling accomplishment.RESULTS:The body weight of AR group and Lung deficiency and AR group reduced and nasal symptoms of them were apparent.The pH volume of nasal secretion and serum IL-2 of AR group and deficiency of Lung and AR group decreased,and their serum IL-4 and Ig-increased with rising of the degree of inflammatory infiltration of nasal septum mucosa and contend of eosinophile granulocyte,the amount of mastocyte of nasal septum mucosa increased especially in the latter group.In the herbal group,pH volume was better that that of AR group and deficiency of Lung Qi and AR group after 7-day medication,serum IL-2 raised and IL-4 and Ig-E lowered,the degree of inflammatory infiltration and contend of eosinophile granulocyte reduced,the amount and function of mastocyte rebounded near the normal volume of control group. CONCLUSION:.The syndrome of traditional Chinese medicine of deficiency of Lung Qi would aggravate the pathological state of AR.The response of experimental AR rats in the state of deficiency of Lung Qi on sensibiligen is stronger than that of AR group that means they are more likely suffering from allergic rhinitis.Herbs of Qi-replenishing and Lung-tonifying influence the experimental AR rats through regulating multiple level and targets such as cytokine and suppressing the synthesis of IgE.
