| Objective: By observing the pathological changes of peripheral facial paralysis rats' facial nerve and the positive neurons of nitric oxide synthase (NOS) in the expression of facial nerve tissue, to contrast the effect of spininess and shallow needling with Spininess and fast Needling in expression of NOS. Discuss the mechanism of acupuncture treatment for peripheral facial paralysis.Method: This experiment totally used 60 peripheral facial paralysis rats which divided into five groups: blank group, false-surgical group, model group, the spininess and shallow needling group, the Spininess and fast Needling group. Use different methods of acupuncture to treat on the rats' faces. Seven days later , we selected the material from the experimental animal, and observed the pathological changes of peripheral facial paralysis rats' facial nerve. Using immunohistochemistry to examine the positive neurons of nitric oxide synthase (NOS) in the expression of facial nerve.Result: 1. We observed the model group with optical microscopy, the main pathological changes are the nerve fibers edema, distorted, narrow, disintegration of the nerve membrane. Under high-power microscope, the continuity of nerve fibers is incomplete, inflammatory cells infiltrated obviously, vacuolar changes in cross-section, small diameter of nerve fibers, thinner myelin. After treatment, the spininess and shallow needling group, the Spininess and fast Needling group are better in nerve injury. The structures of nerve fibers are more normal than model group, the swelling reduced significantly, membranes are more complete.2. Both the spininess and shallow needling group and the Spininess and fast Needling group can inhibit iNOS, activate nNOS, reduce iNOS and enhance nNOS. The two groups have significant differences in iNOS and nNOS. But there is no significant differences in eNOS.Conclusion: 1. Both the two methods can improve facial nerve injury effectively. 2. The spininess and shallow needling is better than the Spininess and fast Needling in the expression of the positive neurons of nitric oxide synthase (NOS) of facial nerve tissue. |
