| ObjectiveAcne is a kind of chronic hair follicles and sebaceous glands inflammatory disease. It is always found on the face,chest and back, where the sebaceous glands secrete sebum highly. The clinical presentation of acne can range from a comedonal from to papule,pustule,node,severe inflammatory cyst, accompanying the overflow of sebum. The sebaceous glands on the bilateral costal regions on the back of golden hamster were taken as the animal model , and study the preventive and therapeutic effect of the complex prescription Qingre Cuochuang tablet(QCT) on acne. In order to explain the functional mechanism of QCT in the treatment of acne, we measure the sebaceous glands, observe the microstructure of the sebaceous glands and checkestrogen and androgen in serum of the golden hamster so that it can be provided as abase of animal experiment for clinical application and furture development.MethodsAll 50 male golden hamsters,weighting 100±20g, were randomly divided into five groups:group I (the model group), group Ⅱ (the Qingre Anchuang tablet group), group III (the small dosage group of QCT) ,group IV (the middle dosage group of QCT) ,group V(the big dosage group of QCT), there were ten golden hamsters in each group. All hamsters were fed with standard feedstuff regularly three days in order to adjust to surroundings,then were irrigated drugs the fourth day. The feather on the back of the hamsers had been cut completely with electric shaver on the beginning.After all hamsters were addicted,we measured the right sebaceous glands by rule on the sunlight. The golden hamsters of each group were fed with 2ml one time a day for 30 days.The hamsters in group I were fed with 0.9% sanile,while the hamsters in groupⅡ ,Ⅲ,Ⅳ,Ⅴ were irrigated with 0.119g/ml Qingre Anchuang tablet liquor,0.828g/ml QCT liquor, 0.414g/ml QCT liquor,0.207mg/ml QCT liquor respectively.After 33 days, we picked off all hamsters' eyeballs and collected their blood samples.Then detected the serum concentrations of sex hormones of hamsters by RIA.Immediately the hamsters were put to death,then measured the right sebaceous glands by rule on the sunlight and took the sebaceous glands on the bilateral costal regions on the back of golden hamster for observing their microstructures under light micscope.Results1 Comparing with group I (the model group), group Ⅴ enable the sebaceous glands of hamsters become smaller(p<0.05).2 Comparing with the model group, group Ⅳ and group Ⅴ can enable the sebaceous glands to become withered,sparse and thin(p<0.01 or 0.05). Group II and group Ⅲ weren't significantly different from group I (p>0.05).3 Comparing with group I ,the level of testosterone(T) of groupⅢ,ⅣandⅤ decreased remarkably(p<0.01 or 0.05) while the testosterone lever of group II wasn't significantly different(p>0.05).The lever of testosterone of group Ⅴ was significantly diffrernt from group II (p<0.05).There is a better dose-effect ralation ship among these groups.4 The level of estradiol 2(E2) of group II ,Ⅲ, Ⅳ and Ⅴ were not significantly different from the model group (p>0.05). The E2 lever of group Ⅴ was not significantly different from the group II (p>0.05).Conclusion1 QCT can enable the sebaceous glands to become small.2 QCT can enable the sebaceous glands to become withered, sparse and thin.3 QCT can remarkably decrease the level of serum testosterone.4 QCT doesn't have an apparent influence on the level of serum estrogen.The results indicate that QCT can remarkably decrease the level of androgen , and influences proliferation of sebaceous glands and inhibits the sebaceous function,reduces the sebaceous glands by regulating the level of estrogen and androgen.It is perhaps the mechanism and theory against acne. QCT is superior to Qingre Anchuang tablet,whose effect is directly proportional to dosage within certain range. |
