Objectives:With traditional Photodynamic therapy with aminolevulinie acid by contrast, research the inhibition effect of of Photodynamic Therapy with the new type of photosensitizer——Pyropheophorbide-a on golden hamster sebaceous patches, for better clinical applications provide experimental basis to treat acne.Methods:(1)The 48male clean level golden hamsters were randomly divided into a:blank control group, b:2.5% ALA-PDT group, c:5% ALA-PDT group, d:2.5% Pheide a-PDT group, e:5% Pheide a-PDT group, f:2.5% Pheide a group, g:5% Pheide a group, h:red light group. A total of eight groups, each group of six. (2)The group b and c topical external using 2.5%,5%ALA to golden hamster sebaceous patches, group d and f topical external using 2.5% Pheide a to golden hamster sebaceous patches, group e and g topical external using 5% Pheide a to golden hamster sebaceous patches,a and h group to apply the same dose of normal saline to golden hamster sebaceous patches.(3)Among these,b, c, d, e group which golden hamster sebaceous patches to be given medicine and keep in dark place 2 hours later, and then red light irradiation. Irradiation power:100 mw/cm2, light wavelength of 635nm, spot size 1.0cm, set the time of 20 min.Photodynamic therapy program was once a week for 4 sessions. (4)Respectively measure the sebaceous patches of the maximum transverse diameter D(T) and the maximum vertical diameter D(v) before and after treatment with vernier caliper,calculating the value of D(v) *D(T) statistical processing.The sebaceous patche specimens were taken for histopathology examination and oil red O staining.Results:(1) Compare with before treatment, the area of sebaceous patches shrink after photodynamic treatment D(T)*D(v) (P< 0.05), of which 2.5%ALA-PDT,5%ALA-PDT significantly reduced(P<0.01). (2) Compare with the blank control group, PDT groups’area of the sebaceous patches D(T)*D(V) were smaller than that of blank group (P<0.05), of which 2.5% ALA-PDT group,5% ALA-PDT group,2.5% Pheide a-PDT group were significantly reduced (P<0.01). (3) Compared with before treatment,medication alone group (2.5% Pheide a and 5% Pheide a) and red light irradiation alone groups’ changes of sebaceous patches D(T)*D(V) have no significantly different(P>0.05).(4) Compared with blank group,medication alone group (2.5% Pheide a and 5% Pheide a) and red light irradiation alone groups’changes of sebaceous patches D(T)*D(v)have no significantly different(P>0.05). (5)Histological observation:blank group has a large number of sebaceous, are intensive arranged multilayer subcutaneous. Compared with the blank group, PDT groups’ sebaceous patches were thin, became smaller, Of which 5% ALA- PDT group and 2.5% Pheide a-PDT group have the highest degree in the sebaceous patches reduce.Sebaceous patches obviously thinning, even disappeared, leaf number of overlapping glands of 0-1, and arranged loosely, scattered in the subcutaneous, liquefied ultricle form. From this, in low dose group (2.5% concentration),Pheide a-PDT has better effect to restrain sebaceous paches, among high dose group (5% concentration) ALA-PDT group’s inhibition effect is better. (5) Compare with the blank control group and medication alone group,2.5% Pheide a group and 5% Pheide a group are similar to the blank group, have no obvious changes, only one case has a slightly reduced in 5% Pheide a group (6) Compare with the blank control group and red light irradiation alone group, most of the sebaceous are similar to the blank group, there are no obvious change, only 2 cases have slightly reduce.Conclusion:(1) the Pheide a-PDT has the obvious inhibitory effect on the sebaceous patches, it can destroy the structure of the sebaceous patches, sebaceous patches area was reduced. Compared with positive control group, low concentration (2.5%) group Pheide a-PDT’s effect is as well as ALA PDT group,high concentration (5%) group ALA-PDT’s effect is better than Pheide a-PDT. (2) simple medication group, the red light group has no obvious inhibitory effect on the sebaceous patches. |
