The research is based on the rational theory of TCM, combined professor academic opinion, researched mechanism and treatment on acne, by animal experiment to explore the mechanism of Acne with the prescription of nourishing-yin and clearing-hot。 reviewed the name, mechanism, treatments on acne; and summarized domestic and abroad latest clinic and animal research on acne; yin deficiency and yang heat was the major factor, heat of lung and stomach was extra-appearance; eat stimulate and hot a lot, rhythm of life changed were the important motivations; sputum and deficiency were the key of acne increased and repeated. Our therapy aims mainly at nourishing Yin and clearing heat. According to this theory, we make up Rehmanniae、Ligustrum lucidum、Yam、Anemarrhena、Treats、Scutellaria、Peony root、Salvia、 Capillaris and Hedyotis diffusa.Objective:The prescription of nourishing-yin and clearing-hot as a classic treatment for acne made a good effect in the clinical, and androgen-related research in acne pathogenesis has been the focus of attention. Based on this, The study is considered the golden hamster of study drug anti-androgen and anti-activity of sebaceous hyperplasia of the ideal animal model for experimental study, observed the changes of sebaceous glands morphology of after the intervention of traditional Chinese medicine, to clarify the mechanism of nourishing-yin and clearing-hot prescription, and to provide the theoretical and scientific basis for treating acne with the prescription, also to provide new ideas in prevention and treatment of acne with Traditional Chinese Medicine.Methods:All42male golden hamsters, weighing130-150g, giving normal breeding, were randomly divided into four groups after a week:blank group had12golden hamsters, the big dosage group of the prescription, the small dosage group of the prescription had10golden hamsters.At the beginning, we cut the bilateral back feather of golden hamsters completely with an electric shaver, exposed bilateral sebaceous glands under bright light. We measured the maximum diameter (DT) and maximum longitudinal diameter (DL) of bilateral plaques with caliper on the sunlight. The hamsters each group were fed with10times drug of people, except the small dosage group that fed with5times drug of people. Animals of the big dosage group were fed with18g/kg crude drug, and the small dosage group were given9g/kg crude drug, and the positive group were10mg/kg dosage. Blank group take amount (Iml/100g) saline. The golden hamsters of each group were fed once per day for4weeks. At the end of the experiment, we shave golden hamsters back hair with an electric razor again, fully revealed bilateral sebaceous glands spot, and measured DT and DL of bilateral sebaceous glands with caliper in the glare. We put the hamsters to death immediately, tooking the golden hamster sebaceous glands organization, and cut off the tissue, putting them in each small bottles filled with10%formaldehyde fixed liquid. Then the conventional paraffin, section, HE dyed, and looked the sebaceous glands for observing their microstructures under light-microscope. Removal of the testes after the Animals were sacrificed, and the removal of visceral fat around the surface of blood, check the weight, balance, calculate the testicular factor (testis weight/body weight×100).Results1. The sebaceous glands size:before and after treatment in each group:Compared with the sebaceous glands before treating, big dosage group and positive group were smaller, which have significant difference (p <0.05)than before. The small dosage group were smaller than before, but have no significant difference (p>0.05).However, Blank group have no significant difference (p>0.05). After treatment between the two groups:Compared with the Blank group, the sebaceous glands of big dosage group and positive group become smaller, and have significant difference (p<0.05). The small dosage group have no significant difference (p>0.05). Compared with the positive group, the sebaceous glands of the big dosage group has no significant difference (p>0.05). The positive group compare with the small dosage group has significant difference (p<0.05).The sebaceous glands of big dosage group and small dosage group have no significant difference compared with each other(p>0.05). The big dosage group of the prescription of nourishing-yin and clearing-hot efficacy similar to positive group. The dose-effect relationship exists between the big dosage group and the small dosage group.2. The sebaceous glands spot microstructure:the big dosage group, small dosage group and positive control group were a blank group loose sebaceous glands, liquefied capsule form number. The thickness of the sebaceous glands comparison:the big dosage group, small dosage group and positive control the thickness of the sebaceous glands are more blank group thin, with no significant difference (p>0.05); the big dosage group, small dosage group sebaceous glands thickness and positive compared has no significant difference (p>0.05). The comparison of the sebaceous glands loose degree:compared with the blank group, the big dosage group and positive control group sebaceous glands with degree of loose have significant difference (p<0.05); the small dosage group have no significant difference (p>0.05). compared with the positive group, the big dosage group have no significant difference (p>0.05), the small dosage group have significant difference (p<0.05).3. Testicular coefficient:Compared all the groups, which have no significant difference (p>0.05).The prescription of nourishing-yin and clearing-hot and spironolactone have no significant effect with golden hamster testis. conclusion1the big dosage group, small dosage group and spironolactone all can make the sebaceous glands spot area is narrowing.2the big dosage group, small dosage group and spironolactone all can make the sebaceous glands atrophy, loose, become variegated thin, which have dose-dependent relationship within certain range.3.This shows, the prescription of nourishing-yin and clearing-hot can inhibit the sebaceous glands cell proliferation and secrete a function, the influence of the sebaceous glands spot hyperplasia, effectively narrowed the sebaceous glands spot, with significant effect on the gonads of experimental animals.
