PartⅠObjective:To investigate effects of Benefiting-bone Granule(BBG) on analgesic experiments and antiphlogistic effect.Methods:The 2 kinds of analgesic experiments of whirling body and hot board were respectively adopted in mice.Results:low dosage,middle dosage and high dosage BBG groups can all inhibit reaction of whirling body by chemical irritation.BBG could inhibit the proliferation of granuloma.Conslusion:BBG has the analgesic effect and antiphlogistic effect.PartⅡThe effect of Benefiting-Bone Granule on microcirculation in miceObjective:To investigate effects of Benefiting-bone Granule(BBG) on microcirculation in mice.Methods:Mice were randomly divided into 5 groups including Control group,GSB group,low dosage,middle dosage and high dosage BBG group.To observe the caliber and flow rate.Results:BBG enlarge the caliber and speed up flow rate.Conslusion:BBG can improve the state of microcirculation. PartⅢStudy on the therapeutic effect of Benefiting-Bone Granule on experimental osteoporosis in ratsObjective:To investigate effects of Benefiting-bone Granule(BBG) on biochemical criterion and bonedensity of primary osteoporosis in rats.Methods:The rat osteoporosis model was induced by ovariectomy.The changes of biochemical criterion in serum(E2,ALP,CT and BGP)and in urine(Ca,and P),and bone density.Effects of treatment group(low,middle and high dosages),the model group,the normal control group and the positive control group were compared.Results:the E2 levels in all three dosage groups were significantly increased,evels significantly decreased,and the bone density also significantly increased, compared with those of the model group.Conslusion:BBG can effectively prevent and cure primary osteoporosis.PartⅣEffect of Benefiting-Bone Granule on OsteoblastsObjective:To investigate the effect of rats sera containing Benefiting-bone Granule (BBG) on the cell proliferation of skull osteoblast in neonatal SD rats and the expression of IL-6 and TNF-αin osteoblasts,and to study the mechanism of BBG treatment for osteoporosis.Methods:Eighty SD rats,aged 12 months,were randomly divided into 4 groups including Control group,low dosage,middle dosage and high dosage BBG group,the sera were obtained at different times.MTT method was used to determine the effect of BBG on osteoblasts proliferation.Osteoblasts were obtained from newborn rats calvaria by collagenase trypsin digestion.Three groups were included in the experiment:Control group,BBG group and CEE3 group.IL-6 and TNF-αwere detected by ELISA.Results:The MTT value reached its highest point 1.5 hours after oral administration(P<0.01).MTT of high dosage BBG met the maximum value with the concentration of 20%(P<0.05).Expression of IL-6 and TNF-αin osteoblasts in the BBG group was decreased compared with that in the Control group(P<0.05),but did not show significant difference from those of CEE3 group.Conslusion:The rats sera containing BBG can promote proliferation of osteoblast.BBG can inhibit the secretion of IL-6 and TNF-αwhich may be one of the mechanisms of action of BBG treatment for osteoporosis.
