Objective:The research was to observe the effects of Benefiting-bone capsule(BBC),a pure Chinese herbal compound preparation,on levels of matrix metalloproteinase-1 and membrane-type matrix metalloproteinase-1 of bone tissue of ovariectomized rats with osteoporosis,and discuss the role of BBC in metabolism of bone matrix in ovariectomized rat.Methods:1.Sixty 10-month-old Sprague-Dawley female rats were used.48 of them were chosen randomly as the model group and made into experimental animal model of osteoporosis with the method of breeding for 12 weeks after bilateral ovariectomy,and the other 12 rats were chosen as the sham-operated group.12 weeks later the bone mineral density of these rats was detected by Dual energy X-ray absorptiometry to assure that the experimental animal model of osteoporosis was replicated successfully.2.Twelve weeks later the rats in the model group were divided into untreated model group, low dose BBC group,middle dose BBC group and high dose BBC group.The rats in the low dose BBC group,middle dose BBC group and high dose BBC group were respectively treated with gastric perfusion of BBC solution that amounted to 1 time,3 times and 9 times dose of BBC used in clinical adult patients,and those in the sham-operated group and untreated model group received gastric perfusion of normal saline,once a day for 12 weeks.3.The change of MMP-1 and MT1-MMP in distal fermur of rats in every group were detected by immunohistochemistry method.4.The differences among groups were compared by ANOVA in SPSS 11.5.Results:1.Twelve weeks after operation,the BMD of different situses in the model group was significantly lower than that of sham-operated group(P<0.05).2.Twenty-four weeks after operation,the expression of MMP-1 of untreated model group were significantly higher than that of sham-operated group(P<0.05);that of low dose BBC group,middle dose BBC group and high dose BBC group were significantly lower than that of untreated model group(P<0.05);that of low dose BBC group and middle dose BBC group were significantly higher than that of sham-operated group and low dose BBC group(P<0.05);there was no significant differences between sham-operated group and high dose BBC group(P>0.05).3.Twenty-four weeks after operation,the expression of MT1-MMP of untreated model group were significantly lower than that of sham-operated group(P<0.05);there were no significant differences between untreated model group and low dose BBC group,middle dose BBC group and high dose BBC group(P>0.05).that of low dose BBC group,middle dose BBC group and high dose BBC group were significantly lower than that of untreated model group(P<0.05).Conclusions:1.Ten-month-old SD female rats had been made successfully into experimental animal model of osteoporosis by breeding for 12 weeks after bilateral ovariectomy.2.The express of MMP-1 increased and the express of MT1-MMP decreased.They have important effect in the destroy of bone matrix collegen,and have take part in the process of osteoporosis.3.BBC can dose-dependent decreasing the levels of MMP-1 in bone tissue and depress the activtion of bone absorption.BBC particpate the degenerate of bone matrix.4.There was no significant change of MT1-MMP in bone tissue in the ovariectomized rat model with osteoporsis.
