DNA sample needs to be amplified by PCR before separated by capillary electrophoresis, and if integrate the capillary electrophoresis with the continuous-flow PCR, it will achieve the miniaturization and integtation of biochemical analysis. Therefor, a continuous-flow PCR chip with optional cycle numbers based on FPGA is designed in this paper, which integrates with the capillary electrophoresis microfluid chip, and it achieves the continuous integration of analysis from PCR amplification to capillary electrophoresis.In this paper, ANSYS finite element analysis software is used to design the heater and temperature sensor model with better heating performance. And using MEMS processing technology fabricates a micro flow-through cihp of glass material and silicon chips integrated Pt membrane heater and temperature sensor. The glass chip and three silicon chips are packaged together to constitute the CF-PCR-CE chip. In order to meet the needs of PCR reaction, the intelligence temperature control system is designed based on FPGA which effectively realizates the temperature control of the three regions, including multi-channel AD sampleing input, temperature curve fitting, PID control algorithm, PWM waveform generator function modules, serial communications, the display and driver of capillary electrophoresis detection curve module.By using the CF-PCR-CE chip system designed in this paper, the plant DNA sample was amplified and detected contrastively by method of gel electrophoresis. The experiment results showed that the sysem can basically achieved the amplifition of DNA sample, and the effect of flow rate and cycle number on amplification was discussed according to the results. Using the chip system, the amplified product was directly sent to the capillary electrophoresis channel on the chip for electrophresis, and it achieved the integration analysis from amplificaiton to capillary electrophoresis.
