Thalassemia is a hereditary hemolytic disease which has high morbidity in the south of Yangtze River in China,high performance capillary electrophoresis(HPCE)is the most conventional method of detection of thalassemia in medical which can make a definite screen of thalassemia by the method of detecting the presence of haemoglobin variant in blood.But most of medical capillary electrophoresis apparatus available in current domestic market are foreign productions,which are bulky and expensive.Therefore,this paper develop a system for thalassemia analysis based on capillary electrophoresis,in order to fill the gaps in the field of domestic capillary electrophoresis instrument development.Firstly,bring forward design requirement and system scheme of capillary electrophoresis design by making principle study of HPCE and combining with the characteristics of thalassemia detection.Then,complete the design of system mechanical structure,system liquid road structure,system light path structure,system hardware circuit structure and system software according to the system scheme.Among them,the design of system mechanical structure and system liquid road structure meet the needs of system framework and system leakproofness.The design of system light path structure meet the needs of specific wavelength detection of hemoglobin in thalassemia samples.In system hardware circuit: programmable power supply circuit meet the needs of power supply of system module;programmable high-voltage power supply circuit meet the needs of 0-12 k V continuous adjustable of system electrophoresis voltage;precision temperature control circuit meet the needs of controlling system electrophoretic temperature 37?;Motion control circuit meet the needs of system automation and n L level sample injection of capillary;detection circuit meet the needs of extremely weak light detection of system.Finally,programme chip driver software using Keil ?Vision5,programme PC interface software using Visual Studio 2015,and taking the system hardware and software circuit to comprehensive debugging.After complete of system platform build,taking system key modules to test,including test of highvoltage power supply module,temperature control module and detection module.After their tests are qualified,carry out tests of system precision and system sensitivity,meanwhile,optimize system electrophoresis voltage,system injection volume,system injection concentration and system light source intensity by repeated tests of quality-controlled samples.After system optimization,using the system to test thalassemia blood samples,including quality-controlled liquid CE test,?-thalassemia blood CE test and ?-thalassemia blood CE test.The experimental results show that the capillary electrophoresis system designed in this paper has the highest sensitivity when the electrophoresis voltage is 9800 V and the sample is diluted 6 times;the system precision is less than 2%,which meets the hemoglobin detection requirement of thalassemia;and the system can successfully analyze typical ?-thalassemia and ?-thalassemia blood samples.
