Chip-based capillary electrophoresis (CE) has attracted considerable attention, due to its fast speed, less sample consumption and high resolution. In the past decade, it has become a powerful analytical method and has been applied to many fields, such as life science, analytical chemistry, etc. CE chip has been investigated in-depth at the aspects of basic theory, machining technology, liquid controlling and detection settings. According these, the two-dimensional capillary electrophoresis chip was brought forward. It couples two different separation mechanisms and has a high analysis capability.The work presented in this thesis consists of three parts. One is the fabrication and DNA sizing analysis of CE chip, the second is the design of the two-dimensional capillary electrophoresis chip, and the third is the study of the two-dimensional capillary electrophoresis chip micro fabrication processes.The crossed-channel CE chip which we have constructed, was made of poly(dimethylsiloxane). By using this chip, the primary research of nucleic acid sizing was conducted. The results demonstrated that DNA fragments can be separated and the average theoretical plate number was up to 16,000. The samples with 10 bp differences in length can be separated and the resolution reached 0.43. The problems in present design are also pointed out and discussed in details. Based on the chip-based capillary electrophoresis we have constructed, we designed a two-dimensional capillary electrophoresis chip. With the theoretical calculation of microfluidic control and the optimization of the channel structure, the two-dimensional CE chip was designed.Finally, the fabrication processing of the two-dimensional CE chip was researched, and the manufacture of the chip was presented.
