Surface plasmon resonance (SPR) biosensor has many advantages such as label-free, real-time detection and high sensitivity. It can offer parameters like dynamic characteristics and concentration of the biomolecular interaction, as well as the biospecific interaction analysis. SPR biosensor earns wide attention and application in Biomedical and Chemical fields, especially in proteomics research and drug discovery. How to ensure the micro sampling with uniform speed and the credible switch between different samples becomes one of the key techniques in SPR biosensor research.Facing the research subject"High-throughput phase detection analyzer for biomolecular interaction", this study purposes to design and manufacture a microfluid system for SPR biosensors to ensure the small quantity and uniform speed of the detection sampling, and offer a two-path parallel detection. It also provides the guidance for subsequent research by accumulating experience. The main contents of this paper are:Firstly, design and manufacture the two-path microfluid system. According to the design, including the flow cell, the sampling micro-channel, micro-valves, the interfacing port, as well as the molding die, explore the manufacture processes like die machining, molding, bonding. Take Polydimethylsiloxane(PDMS) as the main material, carry out a PMMA-PDMS composite microfluid system without inner micro-valves, and a simple microfluid system for verification experiment of the micro-valve.Secondly, design and construct the other parts of the whole sampling system, including the automatic sampling system, the arranging of the flow path for two different sampling processes, a double injection pump system for the pre-experiment and the pneumatic control system. This lays a foundation for the further bio-detection and verification experiment of the micro-valve.Finally, take verification test for the feasibility of Oxygen Plasma Treatment, and the result shows that this method can realize the irreversible bonding between PDMS surfaces reliably.Based on the above works, taking saline as sample to take SPR phase detection experiment by using the microfluid system without micro-valves. The experiment result shows that the PMMA-PDMS composite microfluid system can meet the requirements of basic sampling function and detection for SPR phase detection.
