The detection of Surface Plasma Resonance(SPR)is a new type of sensor technology and compared with the traditional sensing technology the SPR sensors have advantages of label-free detection,real-time detection with the whole reaction process,high sensitivity,fast detection and less samples.Because of these advantages,SPR sensing technology is widely used in life science,food safety,health,environmental testing and other fields.Based on the background,principle and development of SPR,this paper set up a SPR detection system with wavelength modulation(WM-SPR)and test the performance of the system.The WM-SPR has good mechanical stability,high sensitivity and refractive index rate.Then,the paper test the mercury ion and Gram-Negative Bacteria with combining the wavelength-modulation SPR and confocal laser scanning microscopy.Firstly,the paper design a SPR sensor chip with single-strand DNA on the gold film and put the chip into LSCI-SPR.In the tap water,the paper realize the high sensitive detection of mercury ion and the detection limit is as low as 0.01ng/mL.The detection limit is lower than the pollution concentration of mercury ion defined by the U.S environmental protection agency.Secondly,a Gram-Negative Bacteria recognition SPR sensor chip was prepared on the gold film surface using micromolecular drug Polymyxin B Sulfate.The paper test the escherichia coli,pneumobacillus and pseudomonas aeruginosa.The testing results achieve a lower detection concentration and have a good linear relationship within a certain range.In the paper,both of two sensing chip have good selectivity and reach the requirement of experimental test.
