This paper emphasizes on the quantificationaJ analysis of relationship between collagen fiber angle and the pressure it receives.The content of collagen fiber and its distribution outside the cell are closely related with different diseases. The directional distribution of collagen fibers has a very important role in studying the function and self-repair of soft tissue. Application of digital image process technology in the field of medicine has offered an accurate, simple, convenient and rapid mean for the analysis of great deal of medical images, especially for the quantitative analysis of smaller pictures, such as microscopic images.Pressure free collagen fibers in the small intestines are oriented at approximately +30 and - 30 to the longitudinal direction. With the gradual increase of intestinal pressure, causes the distribution of collagen fibers to change. At present some of the techniques used to study the quantificational analysis of collagen fiber distribution such as quantative polarized light microscopy and small angle scattering generally are limited for size of the image and time-consuming, or for the limitation to define orientation angle at 90 degree.Firstly, this paper introduces an applicable method for analyzing two dimension collagen fiber angles and their distribution, and figures out the collagen fiber angles in soft tissue through image process, verifies the relationship between collagen fiber angles and the pressure it receives. Furthermore, this paper also makes some quantrficational research on collagen fiber distribution. A special electron biological microscope was used to obtain a series of animal intestine slice images which are processed under polarized light. Then the images were processed to analyze the relationship between the pixel value at each polarized light angle and the approximate collagen fiber angle. Summarize the statistics to get the relationship between stress condition and fiber angle, and further to evaluate the nexus between intestine pathological change and their stress condition.The collection of images are completed by hand, but the analysis is automatic. This method can also be applied easily in various different soft tissues, such as the section images of ligaments, tendons and cardiac muscle etc., and has better applicability. Through computing the pix value ratio to reckon collagen fiber angle has feasibility and reliability. At the same time, mis paper has also offered a kind of effective method for the research of the large area of collagen fiber distribution.
