Breast cancer is a serious malignant tumor for women, its incidence rate has a upgrade tendency in recent years. Even though, regard to the etiology and its occurrence, development and metastasis of the breast cancer has not yet completely clear. The occurrence of breast cancer is more than one gene involved in multi-steps and multi-stage process of development. There are many gene products to precise regulation in the normal cell growth process. Any one part of a problem can lead to uncontrolled cell growth and thus the proliferation and malignant transformation. It has important significance to study of its occurrence and development in gene and oncogene protein level for the early diagnosis and clinical treatment of breast cancer. In recent yeas, many authors found that proliferating cell nuclear antigen is an important biological parameter to reflect cell multiplication. The synthesis and expression play an important role in tumor cell proliferation and have a closely related to the clinical and pathological features of malignant tumors. VEGF (vascular endothelial growth factor, VEGF) is an important angiogenesis factor, over expressed in many human cancers and related with tumor invasion. VEGF plays an important role in tumor growth and its proliferation that who can stimulate vascular endothelial cell division, proliferation, migration, increase vascular permeability, change extracellular matrix structure, thereby promoting tumor angiogenesis. Epithelial cadherin (epithelial-cadherin, E-cad) is a cell adhesion molecule, Studies have shown that E-cad expression decreased or deactivation in many cancer, and cell adhesion function of E-cad protein mediated was wrong, tumor cells homogeneous adhesion reduced, thus it can easily loss and migration from the original lesion, cause tumor metastasis. It has a significant correlation between E-cad over expression with tumorigenesis, differentiation and invasion.It is inconsistent of PCNA, VEGF and E-cad expression in breast cancer and its relationship with clinical pathological features. There are hasn't been reported that combing detected of PCNA, VEGF and E-cad in the invasive ductal carcinoma of breast. Immunohistochemistry EliVision Plus method was used to examine 80 cases of and 20 cases of fibroadenoma of breast. To analysis the expression of PCNA, VEGF, E-cad in invasive ductal carcinoma (IDC) and the relationship between the expression of PCNA, VEGF and E-cad with clinicopathologic features, we find that:1. PCNA was highly expressed in IDCThe Expression of PCNA confined nucleus, uniform distribution of brown grains in nucleus defined positive. 56 cases of invasive ductal carcinoma and 8 cases of fibroadenoma were positive, the positive rate were 70% and 40% respectively, it's significantly higher than control group (P<0.05). there are positive correlation were found between the expression of PCNA and clinical pathological features (P<0.01), such as Axillary lymph node metastasis,TNM stage, Histological grade, but no significant relationship were found with ages and tumor size (P>0.05).2.The expression of VEGFThe expression of VEGF was mainly found in the cytoplasm of tumor cells. 52 cases of invasive ductal carcinoma and 6 cases of fibroadenoma were positive, the positive rate were 65% and 30% respectively, it's significantly higher than control group (P<0.01). there are positive correlation were found between the expression of VEGF and clinical pathological features (P<0.01), such as Axillary lymph node metastasis (P<0.05), TNM stage (P<0.05), Histological grade(P<0.01), but no significant relationship were found with ages and tumor size (P>0.05).3.The expression of E-cadE-cad localized in the cell membrane. 41 cases of invasive ductal carcinoma and all cases of fibroadenoma were positive, the positive rate were 51.75% and 100% respectively, it's significantly lower than control group (P<0.01). There are negtive correlation were found between the expression of E-cad and clinical pathological features, such as Axillary lymph node metastasis (P<0.05), Histological grade (P<0.01), but no significant relationship were found with ages, tumor size and TNM stage (P>0.05).4. The relationship between expression of PCNA,VEGF and E-cad in invasive ductal carcinoma of breast.Forty-one cases of invasive ductal carcinoma were positive expression of both PCNA and VEGF (52.25%),13 cases were negative expression both PCNA and VEGF (16.25%) . there are obviously positive correlation of the expression of PCNA and VEGF by non-parametric Spearman test (r=0.263, P<0.05). Therty-three cases of invasive ductal carcinoma were positive expression of PCNA, but negative expression of E-cad, 17 cases were positive expression of E-cad, but negative expression of PCNA. there are obviously negative correlation of the expression of PCNA and E-cad by non-parametric Spearman test (r = -0.256,P<0.05). 28 cases were positive expression of VEGF but negative expression of E-cad, 17 cases were positive expression of E-cad but negative expression of VEGF, there are no significant relationship were found between the expression of VEGF and E-cad in invasive ductal carcinoma of breast (r = -0.139, P>0.05).The results indicate that:1. It may important role of over expression of PCNA, VEGF and reduced expression of E-cad in the occurrence and development of invasive ductal carcinoma (IDC) of breast.2. The expression of PCNA and VEGF was positive correlation and the expression of PCNA and E-cad was negative correlation, PCNA, VEGF, E-cad to participate in common in the occurrence and development of invasive ductal carcinoma (IDC) of breast.3. It is an important indicator of PCNA, VEGF and E-cad to assess the invasive ductal carcinoma (IDC) of clinicopathological features. Combined detection of expression of PCNA, VEGF and E-cad was great help to correct evaluation of the malignant degree and potential lymph node metastatic, provide important information of diagnosis and clinical treatment.
