BackgroundOzone has strong oxidation, medical ozone is the mixed gas include ozone which is made of pure oxygen and pure oxygen. Differ from living and industrial used ozone, medical ozone must have high pureness and concentration control. The over high concentration of ozone should injure histiocyte.; the over low concentration ozone is easy to deoxidize by anti-oxide, which can not work its pharmacology action. The usually medical ozone concentration is 20~50μg/ml. Currently, the overseas medical ozone instrument had passed approve by SFDA[SFDA (Ⅰ) 20042540177], which has been used in domestic clinic. The history of medical ozone development has near 50 years, but domestic doctor is strange to it. In Europe and other countries, medical ozone is widely used, such as therapy of glucosuric foot, wound or skin ulcer, stroke, ulcerative colitis, arthritis, protrusion of intervertebral disc, and received sufficient affaim.Bacteria infection is a normal healthy problem by nowdays human being. Antibiotic is the great invention that can counter bacteria. The irrelevant use of antibiotic has already become many society and economy problem. It is anxious for the spread of drug-resistant strain, many kinds of super bacteria has been discovered pepertually, which has impacted the healthy and life of human being seriously.Ozone can efficiently sterilizated or destroyed many kinds of microorganism, including the bacteria, epithyte, insert, HAV, HIIV ect. At present, most scholor recognized that mechanism of ozone sterilization is through biochemistry oxygenation that can impact the substantial exchange of microorganism cell. The material sterilization mechanism might include 3 forms:1.ozone can oxygenate and decompose the glucose oxygenation enzyme;2.ozone can effect bacteria virus directly, destroy endocellular DNA, RNA,decompose the protein, lipide and amylase, which bring on cytolysis and decease, interdict the process of metabolism and reproduce.3. Ozone can infiltrate epicyte, invade cell membrane, enter endocellular, act on ectoblast lipoprotein and inner lipopolysaccharide, change the epicyte permeable, result in cytolysis and apoptosis, act on ectoblast lipoprotein and inner lipopolysaccharide, change the epicyte permeable, result in cytolysis and apoptosis.The sterilization to microorganism is impacted by temperature, time and concentration. Medical ozone afford new choice to sterilizate microorganism by its sufficient, broad spectrum and safe character.Ozone treatment has double-sided nature, and the oxidation and concentrations of ozone were directly proportional. If the concentration is low, the effectiveness is poor. Meanwhile, if the concentration is high, it will produce a strong stimulus to the surrounding tissue, and even lead to tissue and organ damage. With the basic research related to ozone treatment, it has gradually been more and more medical attention and application in ozone treatment. But on which conditions medical ozone will have good effect on a variety of bacteria, and if medical ozone have different effects on different bacteria, or how long medical ozone can kill bacteria and reduce adverse reactions, all the questions are needed to research. The current relevant literature is limitted. In this research, the effect of medical ozone within the scope of the safety of concentrations and time in the common clinical practice on Staphylococcus aureus and Escherichia coli will be investigated, which will provide a preliminary basis clinical application to medical ozone.Objective1 To investigate bactericidal effect of different concentrations of medical ozone on Staphylococcus aureus and Escherichia coli;2 To investigate time of medical ozone to kill bacteria, not only having a good killing effect on the bacteria, but also minimize adverse reactions; 3 To make the basis for the clinical application of medical ozone.MethodStaphylococcus aureus, Escherichia coli bacteria were prepared in sterile saline suspension, respectively. Adjust the amount of bacteria by McNamara bacterial turbidimetric method. Different concentrations and time points of medical ozone were used in bacteria suspended solution, and the bacteria killing rate were calculated. Data were expressed in x±s and were analyzed with SPSS13.0 software. The factorial design factors and treatment group were analyzed by multivariate analysis of variance. Different concentrations or different time of the bactericidal rate in the same group were analyzed by ANOVA, and the differences between any two groups compared using LSD method. P<0.05 as significantly different.Result1. Bactericidal effect of different concentrations of medical ozone on Staphylococcus aureus and Escherichia coliDifferent medical ozone concentration killing rate of Staphylococcus aureus was significantly difference (F= 5278.00, p<0.05). The higher the concentration was, the higher the rate of bacterial killing was. If the concentration of medical ozone was 50μg/ml and used 10min, Staphylococcus aureus was kill 100%.Different medical ozone concentration killing rate of E. coli was significantly difference (F= 3293.26, p<0.05). The higher the concentration was, the higher the rate of bacterial killing was. If the concentration of medical ozone was 50μg/ml and used 10min, E. coli was kill 100%.2. Bactericidal effect of same concentrations of medical ozone on Staphylococcus aureus and Escherichia coliThe killing rate of the same concentration of medical ozone on Staphylococcus aureus and Escherichia coli was significantly difference (F= 142.88,90.93, p<0.05). If the concentration of medical ozone was less than 30μg/ml, the killing rate of Escherichia coli was higher than the rate of Staphylococcus aureus. Meanwhile, when the concentration of medical ozone was risen to 50μg/ml, the kill rate of the two bacteria were all 100%. 3. Bactericidal effect of different time points of medical ozone on Staphylococcus aureus and Escherichia coliIn the same concentration of medical ozone, the killing rate of Staphylococcus aureus and Escherichia coli in 5min and 10min were significantly different. With the time increased, the killing rate of Staphylococcus aureus and Escherichia coli in 10min and 15min were all 100%. If the concentration of medical ozone was reached to 30μ/ml, the killing rate of Staphylococcus aureus and Escherichia coli were all 100% in 10min.Conclusion1. The medical ozone within the safe scope in vitro has a good effect in killing Staphylococcus aureus and E. coli. It suggested that the concentration and times of medical should be increased to enhance bactericidal effect in clinical practice.2. The killing effect of medical ozone on Staphylococcus aureus and Escherichia coli is proportional to the concentration. The higher the concentration is, the better killing effect is.3. In the same concentration of medical ozone, the killing rate of Staphylococcus aureus and Escherichia coli is increased with time increased.4.This study found that if the concentration of medical ozone was less than 30μg/ml, the killing rate of Escherichia coli was higher than the rate of Staphylococcus aureus. It suggested that different bacteria may be sensitivity with different concentration of medical ozone, or different bacteria may have different killing mechanisms. So the different bacteria killing mechanisms is required further study.
