ObjectiveTo study effect on semen viscosity with the therapy Eliminating Phlegm and Nourishing Yin, objectively measure semen viscosity with rotary viscometer, objectively quantify semen indicators of pre-treatment and post-treatment, overcome difficulty of objective measurement on clinical efficacy of Chinese medicine. In order to further reveal the nature of semen hyperviscosity (SHV), promote a quantitative method of semen viscosity with great prospect, and make clear mechanism and target of regulating semen viscosity with Chinese medicine compound.Methods①All related literature about SHV were investigated to make a overview, and find the vacant in previous research.②90 subjects conclude 30 normal fertile men(G1 group), and others were randomized into G2 and G3 groups. G2 were treated with Huainan Huojing Decoction (HHD) for 12 weeks, while G3 with VC, VE, Zinc Selenium Tablets (ZST) andα-chymotrypsin. Semen parameters, semen viscosity, seminal plasma PSA, ACP, Zn, citrate were evaluated before and after treatment.Results①Modern adult male's sperm quality decline obviously. SHV is an important pathogenesis in male infertility. The mechanism of SHV caused by various causes has not been fully elucidated. Numerous studies showed that the possible correlation between secretory function of prostate and SHV. Rotary viscometer can detect semen viscosity objectively, quantitatively, accurately and rapidly. ②Clinical trail shows that all the index indicated no significant differences between G2 and G3 before the trail (P>0.05). But statistically significant differences were observed in sperm density, grade a sperm percentage, grade a & b sperms' percentage, sperm motility, VCL, VSL, VAP, seminal plasma PSA, ACP, Zn, citrate and semen viscosity in G2 and G3 patients before and after the trail, and between G2 group to G3 group after the trail (P<0.05). All the index indicated no significant differences between G2 after the trail and G1 (P>0.05). But statistically significant differences were observed in all the index between G1 to G3 after the trail (P<0.05).ConclusionAfter the therapy Eliminating Phlegm and Nourishing Yin, all the index indicated statistically significant differences to pre-treatment and the control group, including semen quality, semen viscosity, seminal plasma PSA, ACP, Zn, citrate.It can be objective and rapid to measure semen viscosity with rotary viscometer quantitatively. The viscometer has been calibrated by standard viscosity material. Therefore, results achieve high accuracy. Objective measurements of pre-treatment and post-treatment, if integrated with conventional items'analysis, can be significant to analyze etiology and predict prognosis. It can also provide a judgment of efficacy of diagnosis and treatment with an important indicator.All the results show that the therapy Eliminating Phlegm and Nourishing Yin has a good effect on SHV. The therapy can resume the disorder secretory function of prostate and play a positive role in varying degrees on sperm motility parameters. It can improve the penetration of sperm, enhance sperm fertilizing capacity and the birth rate thereby.But SHV seems not to be due to a single mechanism. Other mechanisms and target require further study to better understand their role in SHV.
