Abstract/Summary:
Objective:To research①the association of CD40-E1SNP(-1C/T) gene polymorphism with acute coronary syndrome(ACS) .②whether there is correlation between Serum soluble CD40 ligand (sCD40L) and matrix metalloproteinase -2 (MMP-2)③whether there is the Important effect of CD40-CD40L system in ACS .Method:The CD40-E1SNP(-1C/T) were assayed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) in definite diagnosed 74 acute myocardial infarction patients,92 unstable angina pectorisb patients,and 77 controls with negative results of coronary arteriongraphy.Combined with analyzing the difference of serum inflammatory markers (Solute CD40 Ligands,Matrix Metalloproteinases-2) to reveal effect of CD40-CD40L system in ACS.Results:The CD40-1C allele frequency was 0.655 in AMI group,0.543 in UAP group and 0.455 in controls,the difference has statistical significance(P<0.05).The frequency of CC genotype was 0.419 in AMI group ,0.283 in UAP group and 0.195 in controls, the difference has statistical significance(P<0.05).Compared with controls,the content of serum MMP-2 and sCD40L were higher in UAP and AMI groups,and them in AMI group were higher than in UAP group, the difference has statistical significance(P<0.05). The content between MMP-2 and sCD40L in three groups were linear relationship(P<0.05),and the content of. MMP-2 rose with sCD40L increasing,it has statistical significance(P<0.05).Conclusion:①The CD40-1C/T polymorphism was associated with ACS.②C allele may be the predisposing gene in ACS,and is a factor prone to AMI;The content of sCD40L and MMP-2 in serum stepping up is a signal of unstable atherosclerotic plaque,it may be a prediction for cadiovascular risk events.③CD40-CD40L system is an important role in the development of ACS。...
