Objective:1.To study the correlation between single nucleotide polymorphism and ischemic stroke(IS)in CD40 gene rs3765459.2.Further explore the correlation between single nucleotide polymorphism of CD40 gene rs3765459 and Patients with ischemic stroke were classified according to K-TOAST stroke subtype,gender and age.Methods:A total of 201 patients with ischemic stroke were selected as the case group,and a group of 139 healthy people who were hospitalized at the same time in the hospital were selected as the control group.All the subjects signed the consent form.Genomic DNA samples were extracted from the whole blood with the standard phenol/chloroform protocols.The CD40gene rs3765459 locus A/G polymorphism was genotyped by TaqMan probe method.Differences in the genotype and allele frequencies between two groups were compared by?~2 test.At the same time,the genotype and allele distribution of the two groups were compared according to the subtype of K-TOAST,the gender and age.The relative risk is expressed as OR(95%CI).Results:1.The distribution of G/G genotype and the frequency of G allele in IS patients were significantly higher than those in the controls,and difference between the two groups was statistically significant,G allele increases the risk of ischemic stroke(OR=1.490,95%CI:1.082-2.052).2.Stratification analysis found that,in male subjects,the G/G genotype and G allele frequency of the case group were significantly higher than those in the control group(P<0.05).3.The frequency of G/G genotype and G allele was significantly higher in atherosclerotic cerebral infarction group than other subgroups According to the K-TOAST classification.4.Stratified analysis of age was conducted according to the age of 50,there was no significant difference in allele or the genotype distribution between the case group and the control group.Conclusion:1.CD40 gene rs3765459 locus single nucleotide polymorphisms is associated with the susceptibility of ischemic stroke,and G allele may be associated with increased risk of IS.2.At the same time,the genotype distribution of rs3765459 locus is associated with stroke subtype(atherosclerotic cerebral infarction)and gender(male).
