Background:Graves disease, also known as diffuse goiter,and Hashimoto’s thyroiditis belong to autoimmune thyroid diseases.The incidence of GD in the global is close to 0.5%.As the most common form of autoimmune thyroid disease,the research which emphasize on its pathogenes is more and more. It is considered that the occurrence of GD is releate to variety of factors such as genetic, immunological, environmental fator. Among this,the immune process plays an important role in the development of GD. CD40 plays a key role in the immune response happened in the process of GD. For the beginning of Graves’ disease(GD) hair treatment, the overwhelming majority of domestic physicians preferred ATD therapy, but the prognosis of GD patients in the process of antithyroid drug therapy manifests individual differences,especially withdrawal relapse problem.The correlation between CD40 and GD abroad rarely reported. Objective:By investigating the relationship between the C/T polymorphism in the CD40 gene position-1 site and the relapse of GD after antithyroid withdrawal in Ji Lin Han population,we anticipate to identify whether the early screening GD patients have the CD40-1C / T polymorphism,to provide a strong theoretical basis for Graves’ disease(Graves disease, GD), select the appropriate clinical means of treatment, aviod the pathological damagement of cardiac and other vital organs in the people who recur GD after antithyroid withdrawal,avoid blindness and randomness in clinical drug treatment of Graves’ disease(Graves disease, GD). It has good prospects. Methods:One hundred and sixty-four patients with GD from January 2014 to Dec 2015 in the Second Hospital of Jilin University and One hundred and ten healthy subjects were included.The DNA was extracted from the peripheral blood of these patients and healthy subjects.164 patients,including 80 GD patients without relapse and 84 GD patients Who recur GD after antithyroid withdral,and 110 healthy controls were studied.The genotype and allele frequencies were assayed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).The genotype frequency and the allele frequency were calculated by direct counting. The relationship between the C/T polymorphism in the CD40 gene position-1 site and the relapse of GD after antithyroid withdrawal in Ji Lin Han population were analyed by statistical methods. Rusults:Significant differences were observed in CD40 gene between GD and control group(X2=15.011,P=0.001;X2=8.222 a,P=0.004).The CC genotype frequency was significantly higher in GD group than that in control(23.2%vs13.6%).The TT genotype frequency was significantly lower in GD group than that in control(7.3%vs22.7%). Significant differences were observed in CD40 gene between GD without recurrence and relapse group(X2=10.795 a,P=0.005). The TT genotype frequency in relapse group was significantly lower than that in GD group without recurrence(7.5%vs22.4%).But the Significant differences in allele frequenceies were not observed(X2=0.319 a,P=0.572). Conclusion:The polymorphism of C allele in CD40 gene position-1 site is associated with the susceptibility to the development and relapse of GD in Ji Lin Han population.
