Objective:To investigate the effect of arecoline on the expression and distribution of AngⅡand one of it's receptors-AT1R in Hacat cells and oral mucosal fibroblasts(OMFB),and to further gain insight into the pathogenesis of OSF.Methods:The specific SABC immunocytochemistry was used for detecting the expression of AngⅡand AT1R rabbit anti-human polyclonal antibodies in Hacat cells and OMFB after 48-hour exposure to arecoline of various concertrations.The rate of positive reaction were observed and counted blindly,and the staining intensity was judged by the rate of positive cells.Results:(1) From the microscopic observation,after 48-hour exposure to arecoline of Hacat especially high concertration(80μg/ml),some cells have morphological changes,and these cells in the field of vision is uniformly distributed.(2) Positive expression of AngⅡand AT1R was detected in normal Hacat of the four groups,and the exposure of Hacat to Arecoline at concertrations of 80μg/ml elevated AngⅡand AT1R expression,the difference was statistically significant(p<0.01).(3) Negative expression of AngⅡand AT1R was detected in normal Hacat and the cells exposed to arecoline of 20μg/ml.Slightly positive expression was detected in the cells exposed to arecoline of 40μg/ml. Positive expression was detected in Hacat exposed to areciline of 80μg/ml.The difference was statistically significant(p<0.01).(4) The expression of AngⅡand AT1R was detected frequently in the cells which have morphological changes.(5) Negative expression of AngⅡand AT1R was detected in all the four groups of OMFB,and there is no morphological change in OMFB exposed to arecoline of 20-80μg/ml for 48 hours.Conclusions:(1) Arecoline at a certain concertration can upregulate AngⅡand AT1R expression in Hacat,and the expression of these two protein was detected frequently in the cells which have morphological changes.These indicate that AngⅡand AT1R may contribute to the pathogenesis of OSF.(2) Arecoline of 20-80μg/ml can not change the expression of AngⅡand AT1R in OMFB,and there is no morphological change.
