The genus Triosteum Linn belongs to the family Caprifoliaceae,and has about 7 species distributed from Central to East Asia and North America,but there are only 3 species distributed in China.Up to now,no phytochemical constituents of them have been studied.Triosteum himalayanum Wall,a species of Triosteum Linn,has been traditionally applied for many medical purposes,including inducing diuresis and promoting blood circulation,and distributed particularly in Hebei,Shanxi,Shaanxi,Ningxia,Gansu, Qinghai,Henan,Hubei and Sichuan in China.In order to explore more plants with medical value in Triosteum,and develop the new herbal plant resources in China,we carried out systemic phytochemical studies on the whole plant of T.himalayanum Wall,which was collected in Tanchang,a city of Gansu Province.This is the first time to report the phytochemical study on it.From the acetone and methanol extract of the roots,and the methanol extract of the aerial part,37 compounds were isolated by means of silica gel,Sephadex LH-20,D101, Diaion HP-20,MCI gel CHP20P column chromatography,preparative thin layer chromatography,MPLC,and recrystallization etc.The structures of 35 compounds were elucidated by spectroscopic methods(1D,2D NMR,MS,IR,UV,X-ray crystallography),in combination with comparison to the literature data or authentic samples.They were 14 iridoids(including 8 iridoid glycosides and one of which is monoterpenoid indole alkaloid),4 flavonoids,1 monoterpenoid macrolide diglycoside, 2 triterpenes,5 sterols,4 aromatic derivatives,1 coumarin and the others.Among which,four new iridoids were elucidated as triohima A(1),triohima C(3),4', 6'-O-isopropylidene sweroside(9),secologanin diethylacetal(12).The absolute configuration of novel iridoids 1 and 3 with unusual skeleton was established by comparison the experimental and calculated optical rotation using Gaussian 98 package.Some compounds were tested for the in vitro cytotoxicity and anti plant pathogenic fungus activity. Finally,the chemical constituents and pharmacological activities reports of Caprifoliaceae from 1990 to 2008 were reviewed in this thesis.The results of this thesis would contribute to the research on the chemical constituents of T.himalayanum and supply scientific evidences for searching new natural medicines.This study may also help utilize and develop the natural resources of our country.
