Calophyllum inophyllum L. belongs to Calophyllum genus (Guttiferae). Plants of the Calophyllum genus are known as a rich source of secondary metabolites such as coumarins, xanthones, flavonoids, chromenes, triterpenes etc.The chemical constituents of 95% EtOH extracts from the stems and leaves of C. inophyllum were studied systematically. Twenty-two compounds were isolated by repeated silica gel, sephadex LH-20 column chromatography and prep. TLC. On the basis of spectral data and measurement of physicochemical characters, the structures of these compounds were identified as fiiedelin (1), friedelan-3β-ol (2), canophyllal (3), canophyllol (4), canophyllic acid (5), canophyllalic acid (6), 3β, 23-epoxy-friedelan-28-oic acid (7), oleanolic acid (8), 2-hydroxyxanthone (9), 4-hydroxyxanthone (10), 1, 5-dihydroxyxanthone (11), 1, 7-dihydroxyxanthone (12), 1, 3, 5-trihydroxy-2-methoxyxanthone (13), 6-deoxyjacareubin (14), amentoflavone (15), kaempferol-3-O-α-L-rhamnoside (16), quercitrin (17), calophyllic acid and isocalophyllic acid (18), 12-methoxyinophyllum D (19), gallic acid (20), protocatechuic acid (21), daucosterol (22). Among them, compound 7 was a new compound, compounds 6, 16 and 17 were obtained for the first time from Calophyllum genus and compounds 9-10, 12-14 were obtained for the first time from C. inophyllum.Antiproliferative activities of compounds 5-8 in human HL-60 leukemia cells were tested by growth inhibition method. All these compounds showed inhibitory activity and compound 6 was the most effective antiproliferative agent with an IG50 value of 2.67μM.
