Objective:The purpose of our study was to analyzed the correlation between hypertensive disorder complicating pregnancy and renin-angiotensin system (RAS) gene polymorphisms (T174M and A1166C and ACE I/D ) of placentas of pregnant women.Methods:. The study included 55 patients with preeclampsia and 70 placentas of normal pregnant women. Screening for the RAS gene polymorphisms (T174M and A1166C and ACE I/D) was carried out by analysis of polymerase chain reaction–restriction fragment length polymorphism. Statistical significance was regarded as p<0.05.Results:1. In the research of T174M polymorphism:In the research of all the samplesT174M polymorphism, we found there were no statistically significant differences for the frequency of genotype and the T allele of AGT genes between preeclampsia group and the normal group, and there were no TTgenotype of all subjects .2. In the research of A1166Cpolymorphism:(1) We found that the constituent ratio of AT1R genotype between the preeclampsia group and the normal group had statistical significance (Fisher exact probability was 0.028,P <0.05), that was to say, the frequency of theA1166C variant AC genotype was significantly higher in the preeclampsia group (18.2%)than in the control group(5.7). In addition, the C allele frequency was significantly higher in the preeclampsia group (9.1%) than that in the control group (2.9%), too. there were statistically significant differences for the frequency of allele of AT1R gene between the preeclampsia group and the normal group, (P=0.033<0.05).(2)We used logistic regression models to estimate the odds ratios of risk for preeclampsia associated with AT1R gene polymorphism . Compared with AA genotype,there was a significant relationship between AC genotype of AT1R gene preeclampsia (OR; 3.667, 95% CI; 1.083~12.418, p=0.028) .The frequency of the C allele of the AT1R polymorphism plus A allele was significantly higher in pre-eclamptic patients than in controls (OR=3.4, 95% CI;1.036~11.153;p=0.033).3. In the research of ACE I/Dpolymorphism:(1)We found that the constituent ratio of ACE genotype between the preeclampsia group and the normal group had statistical significance (Fisher exact probability was 0.046,P <0.05), The percentage of the ACE DD/DI/II genotype were respectively as bellows:21.8%/52.7%/25.5% and 11.4%/57.2%/31.4%. In addition, the D allele frequency was significantly higher in the preeclampsia group (48.2%) than that in the control group (32.9%), too. there were statistically significant differences for the frequency of allele of ACE genes between the preeclampsia group and the normal group,(P=0.014<0.05).(2)We used logistic regression models to estimate the odds ratios of risk for preeclampsia associated with ACE gene polymorphism . compared with II genotype,there was a significant relationship between DD genotype of ACE gene of preeclampsia (OR; 3.429,95% CI; 1.008~8.282, p=0.024) .The frequency of the D allele of the ACE polymorphism plus A allele was significantly higher in pre-eclamptic patients than in controls (OR=1.9, 95% CI;1.136~3.177,P=0.014).4. Combination of angiotensin-converting enzyme I/D and AT1R A1166C polymorphismsWe used logistic regression models to estimate the odds ratios of risk for preeclampsia associated with ACE and AT1R gene polymorphisms. compared with II-AA genotype combined,there was a significant relationship between DI-AC and DD-AC genotype combined of preeclampsia . (OR;9.545,95% CI; 1.715~53.127, Fisher exact probability was 0.007) .Conclusion:1. We concluded that the T174M gene in the jiangxiHan placentas didn't find correlation to preeclampsia2.We conclude that the placentas's AT1R gene A1166C polymorphism may have correlation to preeclampsia, the presence of the A1166C gene in placentas could be a marker of increased risk of developing preeclampsia.3. We conclude that the placentas's ACE gene I/D polymorphism may have correlation to preeclampsia ,the presence of the ACE gene in placentas could be a marker of increased risk of developing preeclampsia.4. Synergism of angiotensin-converting enzyme I/D and AT1R A1166C polymorphisms of renin-angiotensin system may be a risk factor for preeclampsia.
