Objective:To analyze the correlation between hypertensive disorder complicating pregnancy and the Methylenetetrahydrofolate reductase(MTHFR)gene polymorphism (Glu429Ala) of pregnant women and their fetus.Methods:The sample of this study include 64 women with hypertensive disorder complicating pregnancy (20 patients with gestational hypertension, 44 patients with preeclampsia) and 62 normal pregnant women,68 placentas of the women with hypertensive disorder complicating pregnancy(17 patients with gestational hypertension, 51 patients with preeclampsia)and 46 placentas of normal pregnant women at the same time. The assay method for the screen of MTHFR gene polymorphisms is polymerase chain reaction–restriction fragment length polymorphism(PCR-RFLP).Results:In the research of all the samples about Glu429Ala polymorphism, we found that :①In the research of mother's genotype: the constituent ratio of AC,CC genotype in the hypertensive disorder complicating pregnancy group is not different from that in the normal group; at the same time, the frequency of C allele in the hypertensive disorder complicating pregnancy group is not different from that in the normal group. Divide the hypertensive disorder complicating pregnancy group into two groups: gestational hypertension group and preeclampsia group, and compare the two groups with control group individually,We have the same result.②In the research of fetus's genotype: the constituent ratio of AC,CC genotype between the hypertensive disorder complicating pregnancy group and the normal group had no statistical significance .But, the frequency of C allele in the hypertensive disorder complicating pregnancy group is higher than that in the control group (P <0.05). Divided the hypertensive disorder complicating pregnancy group into two groups: gestational hypertension group and preeclampsia group, and compared the two groups with control group individually. We found that the constituent ratio of AC,CC genotype between the gestational hypertension group and the normal group had no statistical significance , at the same time , we got the same result between the normal group and the preeclampsia group; we found that the frequency of the C is not different between the gestational hypertension group and the control group. But, the frequency of C allele is higher in the preeclampsia group than that in the control group(P <0.05).Conclusion:We conclude that there is not the correlation between mother's MTHFR gene Glu429Ala polymorphism and hypertensive disorder complicating pregnancy. On the contrary, the fetus's MTHFR gene Glu429Ala polymorphism may have correlation to hypertensive disorder complicating pregnancy. But it is preeclampsia not gestational hypertension that has correlation to Glu429Ala polymorphism. So, the presence of the Glu429Ala gene in fetus may be a marker for the risk of preeclampsia.
