Objective:Our experiment was to discover the differences of Hypoxia-inducible factor-1α(HIF-1α)and Aspartyl-(asparaginyl) beta-hydroxylase(AAH)expression level in villi between missed abortion patients and normal women with early pregnancy,and confirm the expression loci of HIF-1αand AAH in villi.Methods:50 patients of missed abortion were collected and categorized into test group(from Feb 2007 to Dec 2007,they were all diagnosed by B-ultrasound andβ-HCG continuous monitoring),which was subdivided into group 1 and group 2.Patients in group 2(n=30) showed no obviously etiological clues while those in group 1(n=20) were of confirmed etiological disorders.In addition,30 women of early pregnancy with artificial abortion surgery were categorized into control group,whose embryos were sonographically confirmed alive before surgery.50 patients of missed abortion were also subdivided into group within 4 weeks and group excess 4 weeks according to the time that the embryo stayed in utrine after died.Immunohistochemical technique and computer image analysis were used to detect the expression loci and level of HIF-1αand AAH in villi.Results:1.HIF-1αexpression was detected both in missed abortion villi(test group)and normal early pregnancy villi(control group).The expression location was in endochylema and nucelus of trophocyte.The level of HIF-1αin villi of group 1(average gray scale115.7300±8.3898)was lower than in that of control group(average gray scale84.4950±11.3814) (P<0.05).The level of HIF-1αin villi of group 2(average gray scale117.3400±11.4687)was lower than in that of control group(average gray scale84.4950±11.3814)(P<0.05),and there was no significant difference of HIF-1αexpression level between group1(average gray scale115.7300±8.3898)and group 2(average gray scale117.3400±11.4687).There was no significant difference of HIF-1αexpression level between within and excess 4weeks that the embryo stayed in urine after died.2.AAH expression was detected both in missed abortion villi(test group)and normal early pregnany villi(control group).The expression location was in endochylema and nucelus of trophocyte.The level of AAH in villi of group 1(average gray scale149.000±18.7083)was lower than in that of control group(average gray scale124.5000±12.0372)(P<0.05).The level of AAH in villi of group2(average gray scale150.0333±17.7015)was lower than in that of control group(average gray scale124.5000±12.0372)(P<0.05),and there was no significant difference of AAH expression level between group1(average gray scale149.000±18.7083)and group 2(average gray scale150.0333±17.7015),either.There was no significant difference of AAH expression level between within and excess 4weeks that the embryo stayed in urine after died.Conclusions:1.The expression loccation of HIF-1αis in endochylema and nucleus of trephocyte both in missed abortion and normal early pregnancy.The expression level of HIF-1αin villi of missed abortion is much lower than that of in villi of normal early pregnany.2.The expression location of AAH is in endochylema and nucleus of trephocyte both in missed abortion and normal early pregnancy.The expression level of AAH in villi of missed abortion is much lower than that of in villi of normal early pregnancy. 3.HIF-1αand AAH have a function of supporting normal pregnancy, disequilibrium of expression can result in missed abortion.
