Objective:To measure the HGF and TGF-β1serum concentration and theirexpression intensity in the chorionic villi in missed abortion patient and normal earlypregnant women in this study,in order to discuss the relationship of missed abortion andthe factors above.Methods:Enzyme-linked immunosorbent assay was used to detect the HGF andTGF-β1concentration in serum from33missed abortion patients(missed abortion group)and45normal pregnant women (control group) who required for artificial abortion;Immuno-histochemically technique was used to determine the location and intensity ofHGF and TGF-β1in the chorionic villi from26missed abortion patients and44normalpregnant women,and the results were analyzed by the method of semi-quantitativeway.These people are randomly collected form July2012to Novermber2012at theObstetrics and Gynecology department of the subsidiary hospital of Yan’anUniversity.The missed abortion patients satisfied the diagnostic criteria in the7thGynecology and Obstetrics edited by Le Jie,and the two groups have no statisticaldifference in days of pregnancy and age.Results:1The serum concentration of HGF and TGF-β1in the two groupsThe average serum concentration of HGF in the missed abortion group is214.63±62.26pg/ml, and the control group is219.73±49.49pg/ml, the two groups haveno statistical difference (P>0.05).The average serum concentration of TGF-β1in themissed abortion group is (6.40±0.89)×103pg/ml, and the control group is(7.22±0.92)×103pg/ml, the two groups have no statistical difference (P>0.05). There is no correlation between the serum concentration of HGF and TGF-β1(r=-0.116,P>0.05).2The expression in the villous of HGF and TGF-β1in the two groupsHGF is mostly expressed in the cytoplasm of the sertoli cell, the mesenchymal cell,and the vascular endothelial cell.TGF-β1is mainly expressed in the cytoplasm of thesertoli cell and the mesenchymal cell.In the missed abortion group, the mean rank of theexpression intensity of HGF is significantly lower compared with the controlgroup(Z=-4.634,P<0.05),while there is no statistical difference in the expressionintensity of TGF-β1between the two groups(Z=-1.364,P>0.05).There is no statisticalcorrelation between the expression intensity of HGF and TGF-β1in the same villi(rs=0.459,P <0.05).Conclusion:1The serum concentration of HGF and TGF-β1in the two groups have no statisticaldifference, this indicates that the serum level of HGF and TGF-β1has no correlation withmissed abortion.2HGF is expressed in the villous of the two groups, its expression in the missedabortion people is statistically lower.It indicates that the normal expression of HGF maytake a part in the normal pregnancy, and its lower expression may be associated with theoccurrence and development of missed abortion, the decline of HGF expression maydecrease the trophoblastic proliferation, increase apoptosis, deline the invision ability bya variety of mechanisms, and affect the cultivation and development of embryo,eventually to the missed abortion.3TGF-β1is expressed in the villous of the two groups, its expression in the twogroups have no statistical difference.While in the same villous, the expression of HGFand TGF-β1is significantly correlated. It indicates TGF-β1may participate in thedevelopment of missed abortion through regulating the expression of HGF.
