Objective: To investigate the Rosa laevigata effection on the oxidative stress of diabetic nephropathy rats, simultaneously approaching the mechanism of action.Methods: The SD rats induced into diabetic by STZ were divided into the diabetic model groups and the Rosa Laevigata intervention groups, simultaneously setting up the normal control groups and the Rosa laevigata control groups. Blood sugar, kidney weight, TC, TG, BUN, the ability of the T-AOC, the activity of SOD, the amount of MDA in nephridial tissue were determined. the morphological pathological changes in nephridial tissue were Observed. the expressions of NF-κb, MCP-1 and TNF-αin the four groups were detected by immunohistochemistrcey, Western blotting and Realtime-PCR.Results: Rosa laevigata could reduce Blood glucose, index number of kidney, TC, TG, of diabetic rat, and impove renal function. After the intervention of Rosa Laevigata, the pathological changed of nephridial tissue lessen, basal lamina of party of blood vessel thicken, matrix of glomeruli renis and account of mesangial cells decrease obviously, PAS red dyed domain enlarged lightly, mesangial region widen lightly, lumina of blood capillary were compressed lightly. The ability of the T-AOC and the activity of SOD were increased significantly by the Rosa Laevigata and the amount of MDA decreased (P<0.05). Immunohistochemistry image quantitative analysis manifestd that the expressions of protein of NF-κB, MCP-1 and TNF-αin the diabetic model groups enhanced notablely, however, after intervention, the expression attenuated notablely(P<0.05). Western boltting analysis disclosed that the expressions of MCP-1 were decreased from 64.3% to 20.8% and NF-κB were done from 41.2% to 22.3% by the Rosa Laevigata. The outcome Of real time fluorescent quantitation PCR displayed that the amount expression of NF-κB, MCP-1, TNF-αmRNA in the nephridial tissue of the diabetic model groups increased in 2.74, 2.62, 2.89 times than the normal control groups, however, in the Rosa laevigata intervention groups, the expression of NF-κB, MCP-1, TNF-αmRNA decreased in 37.31%, 35.61%, 31.95% than in the diabetic model groups. Conclusions:1. Diabetic rat display that oxidative stress strengthen, peroxidize substance increase, enzymatic activity of antioxidase decrease, the expressions of protein of NF-Κb, MCP-1 and TNF-αincrease.2. Rosa Laevigata can abate oxidative damage of diabetic nephropathy rat, reduce peroxidize substance, heighten enzymatic activity of antioxidase.3. The mechanism that Rosa Laevigata protect the kidney of diabetic rat may involve in the intervention of Rosa Laevigata on the oxidative stress, the suppression of the expressions of protein of NF-κB, MCP-1and TNF-α.
