Objective:To explore traditional Chinese medicine Rosa Laevigata Michx extract liquid on the treatment of experimental IgA nephropathy,understanding its mechanism.Methods:36 rats were randomly divided into normal group,model group and RLM treat group,then IgA nephropathy model were constructed.After treated few weeks by RLM,the rats' proteinuria,the renal pathology and the levels of TXB2 and 6K-PGF1awere examined.To explore the protective mechanism of RLM from gene and protein levels,expressions of MCP-1mRNA and TGF-βmRNA in renal tissues by RT-PCR methods were performed,as well as investigated the protein changes by SDS-PAGE and SELDI-TOF-MS methods.Results:When finished IgA nephropathy model,urine protein of normal group,model group and RLM treat group were 10.77±1.65,43.13±8.78,42.47±8.20,after treated five weeks by RLM extract liquid,they were 8.29±1.31, 45.31±5.33 and 14.90±2.05.Compared with model group,urine protein in RLM treat group decreased significantly(P<0.05).The BUN,Cr,TC,TG were lower in RLM treat group than that of in model group(P<0.05).Light microscopic examination of renal tissue,more mesangial cell and mesenterium groundmass and wider mesangial region existed in model group than in normal group.RLM treat group than the model group degree ease.Detected the contents of TXB2 and 6K-PGF1ain three groups by radio-immunoassay methods,the ratios of TXB2/6K-PGF1ain RLM treat group was 0.71±0.05,which was lower than in model group of 1.444±0.11(P<0.05).In protein aspect,there were two distinct straps in SDS-PAGE.Then further detected by SELDI-TOF-MS,Mw 12164Da of protein was found.It was high expression in normal group and RLM treat group,but low expression in model group.In gene level,relative amount of TGF-βmRNA in three groups were 0.5416±0.1847,1.5022±0.7343, 0.6440±0.0518,and MCP-1mRNA were 0.3083±0.1594,0.8732±0.5017, 0.4182±0.1933.The expressions of TGF-βmRNA and MCP-1mRNA were significantly lower in RLM treat group than in model group(P<0.05).Conclusion:RLM liquid could relieve the protein urine symptoms,cut down BUN,Cr,TC,TG and reduce blood-fat.It was able to balance TXB2/6K-PGF1ain effect,which improve renal blood flow and regulate the expression of TGF-βmRNA and MCP-lmRNA,in order to relieve inflammation in nephridial tissue.Significantly different protein was detected by SDS-PAGE and SELDI-TOF-MS methods. Objective:To study the effect of Rosa Laevigata Michx liquid on antilipoperoxidation in vitro.Methods:MDA was producted after lipid peroxidation.The rat liver homogenate was affected by different concentration of RLM.Malondial dehyde (MDA)content in vitro was determined by TBA spectrophotometric method.Results:MDA content of the control group was 39.04±0.33,it was suggest that rat liver homogenate had been emerged lipoperoxidation.But when different content(25,50,100,200,400g/l)of RLM deal with it,produces of MDA were 31.08±0.49,30.40±0.45,29.46±0.36,24.34±0.41,20.95±0.32, respectively.RLM could inhibit lipid peroxidation in rat liver homogenate,and there was a dose-response effect.Conclusion:Rosa Laevigata Michx could inhibit lipid peroxidation and has antioxidation effects in a range of certain concentration.
