Objective:The root of Rosa laevigata Michx.?R. laevigata.? have the effects of astringent, eliminating poison, promoting blood circulation to remove blood stasis, expellingthewind, which is the main components of SanJinpian, JinJiJiaonang, FukeqianJinpian, WanglaoJi, etc. However, the reports of the chemical constituents from the roots of R. laevigata is insufficient and the pharmacological activity is single and uncomprehensive, which needs further research. This article we will systematic study the chemical composition of the root of R. laevigata and the anti-inflammatory activity of parts of triterpenoids. Hoping the findings establish chemical basis for the development and utilization of Rosa laevigata Michx.Methods:The petroleum ether, CH2C12, CH3COOC2H5, CH3COCH3, 50% CH3OH were used to extract the chemical constituents from the roots of R. laevigata. The silica gel column chromatography, LH-20 column, D101 Macroporous resin column, HPLC, recrystallization were used to separate and purify the constituents. Physical and chemical properties,1H-NMR,13C-NMR, 2D-NMR, ESI-MS, Optical rotations, UV spectra and IR spectra were used to determine their structures. The effects of triterpenes on the production of NO in LPS-stimulated mouse RAW 264.7 cells was used to study the anti-inflammatory activity of parts of triterpenoids.Results:Forty constituents were isolated and identified as ?-sitosterol?1?, niga-ichigosidesF2?2?, rosamultin?3?, ArJunetin?4?, kaJi-ichigoside Fi?5?,12, 13-dihydromicromeric acid?6?, ?2R,19R? ethyl-acetyloxy-9-hydroxyl-3-oxo-urs-12-en-28-carboxylate?7?, pomonic acid?8?, cecropiacic acid 3-methyl ester?9?,2-acetyl tormentic acid?10?, pomolic acid?11?,2a, 3?-dihydroxyurs-12,18-dien-28-oic acid?12?,3?-E-feruloyl corosolic acid?13?, Fupenzic acid?14?,2-O-acetyl euscaphic acid ?15?, Euscaphic acid?16?,18,19-seco,2a,3?-dihydroxy-19-oxo-urs-11,13?18?-dien-28-oic acid?17?, swinhoeic acid?18?, arJunic acid?19?, myrianthic acid?20?,2a,3?, 19?-trihydroxy-24-oxo-urs-12-en-oic acid?21?, Tormentic acid?22?, 1?-hydroxy euscaphic acid ?23?, quercilicoside A?24?,2?,3?, 23-trihydroxyurs-12,18-dien-28-oic acid 28-O-?-D-glucopyranoside?25?, 2a,3a,23-trihydroxyurs-12,19?29?-dien-28-oic acid ?-D-glucopyranoside ?26?, quadranoside ??27?,23-hydroxy-tormentic acid?28?,2a,3?,19a,23-tetrahydroxy-12-ene-28-oleanolic acid?29?,2a,3?,24-trihydroxylup-20?29?-en-28-oic acid 28-O-?-D-glucopyranoside?30?, alpinoside?31?,2?,3?, 24-trihydroxyurs-12,18-dien-28-oic acid 28-O-?-D-glucopyranoside ?32?, Rubuside B?33?,2?,3?,20?-trihydroxyurs-13 ?18?-en-28-oic-acid?34?,2?,3?, 20?-trihydroxyurs-13 ?18?-en-28-oic-acid ?35?,2?,3?,24-trihydroxyurs-12, 19-dien-oic acid 28-O-?-D-glucopyranoside?36?,2?,3?,20?,23-tetrahyd-roxyurs-13?18?-en-28-oic-acid?37?,2?,3?,20?,23-tetrahydroxyurs-13?18?-en-28-oic-acid?38?,1?,2?,3?,20?-tetrahydroxyurs-13?18?-en-28-oic-acid ?39?,2?,3?,20?,24-tetrahydroxyurs-13?18?-en-28-oic-acid?40?. Including five new compounds, one steroid and 39 triterpenoids. The main types of the triterpenoids include ursane, oleanane,lupine. Compounds 2,3,4,9 have a better anti-inflammatory activity.Conclusion:compunds 36-40 were assigned as new compounds, compounds 6-10,12-15,17,18,21,23,25,30-35 are obtained from this plant for the first time. Compounds 6,13,15,17,18,21,25,30-35 are obtained from the genus Rosa. for the first time. Parts of the triterpenoids have the activity of anti-inflammatory.
