Rosa laevigata Michx.(R. laevigata) is a very interesting shrub in the Rosa genus of Rosaceae family. They distribute widely, mainly grow in East China, South-middle China and Southwest China. The fruits of this plant are used for the treatment of diureses and kidney deficiency; the leaves are used as drug of detoxification and detumescence; the roots have the effects of astringent, eliminating poison, promoting blood circulation to remove blood stasis, expellingthewind. Study on chemical constituents from fruits or leaves of R. laevigata has shown there are polysaccharides, flavonoids, steroids and steroid saponins, triterpenoids and triterpenoid saponins, lignans, hydrolysable tannins and some other constituents in R. laevigata, which have well antioxidant effects, protective effects on kidney and hepar, as well as antibacterial and antiviral action, enhancing immunity and anti-inflammatory on body, lowers blood glucose and blood fat and other pharmacological function.However, the chemical constituents from the roots of R. laevigata as traditional Chinese medicine have not been reported so far. In this work we have mainly investigated the chemical constituents from the roots of R.laevigata. Through a variety of methods and techniques of extraction, separation and purification, such as recrystallization, D101macroporous resin column, Saphadex LH-20chromatography, silica gel chromatography, MCI chromatography, ODS chromatography, semi-preparation HPLC and others, based on physical and chemical properties,1H-NMR,13C-NMR,DEPT,2D-NMR,ESI-MS, Optical rotations, UV spectra and IR spectra as well as HR-ESI-MS,the structures of36isolated compounds from85%EtOH extraction from the roots of R. laevigata.were finally determined. Listed respectively as following:13flavanols, containing (+)-catechin (J1), gallocatechin (J2),3,4,7,4’-tetrahydroxyflavane(J3),4-methoxy-3,7,4’-trihydroxy-flavane (J4),8-acetic acid-catechin (J5), procyanidin B3(J6), Fisetinidol (4a'8)-catechin (J7), guibourtinidol-(4β'8)-catechin(J8), guibourtinidol-(4α'8)-catechin(J9), Fisetinidol (4β'8)-catechin (J10), epiguibourtinidol-(4α'8)-catechin (J11), A dehydro-dicatechin A (J12), ent-fisetinidol-(4β'6)-catechin (J13); one chalcone, phloridzin (J14); one stilbene and one stilbene glycoside,(Z)-3-methoxy-5-hydroxyl-stilbene (J15),(Z)-Piceid (J16); three triterpenoids and four triterpenoid saponins, euscaphic acid (J17), tormentic acid (J18), betulinic acid (J19), kajiichigosie F1(J20), rosamultin (J21), Nigaichigoside F2(J22), Rubuside J (J23);one steroid and one steroid saponin, β-sitosterol (J24), sitosterol-3-O-β-D-glucopyranoside (J25); two phenolic glycosides,4-O-β-D-glucosybenzoic acid (J26) and5-hydroxy-3-methoxy-phenyl1-O-β-D-glucopyranoside (J27); three fatty acids, linoleic acid (J28), methyl linoleate (J29) and methyl oleate (J30); six other compounds, glycerol (J31), Et-β-D-gluco-pyr-anose (J32), glucose (J33), gallic acid (J34),(6R,9S)-3-Oxo-a-ionol (6R,9R)-3-Oxo-a-ionol-β-D-glucopyranoside-D-glucopyranoside (J35),(1S)-cembrene (J36).Compounds J4, J5, Jll were identified as new compounds, and cmpounds J2、 J3、J7、J8、J9、J10、J12、J13、J14、J15、J16、J19、J22、J26、J27、J28、 J29、J30、J31、J32、J34、J35、J36were firstly reported from Rosa laevigata.Above research on chemical constituents from the roots of R.laevigata reveals its mainly chemical composition, which establishes basis for studying properties of the roots of R. laevigata as traditional Chinese medicine, improving their curative effect,searching for new medicines, new drug resources and exploiting valuable resources.
