Objective: To study the effect of Traditional Tibetan Drug SibiraeaAngustata extracts on suppressing S180 grow in body and the contents ofIL-2 and TNF-a that spleen cell secrete in mice with tumor , and the effecton the expression of P53,Bcl-2 protein. It will present scientific theoryevidence to investigate and use this Chinese traditional medicine.Methods: Sixty kunming mice were randomly allocated to an six group. Thereare ten mice in each group. 1)Normal control group:the mice were perfusedorally into stomach with nomal saline per every day and nottransplanted tumor cell;2) Model control group: the mice weretransplanted tumor cell and perfused orally into stomach with 0.2mlnomal saline per every day;3)High dose group(6g/ml): the mice weretransplanted tumor cell and perfused orally into stomach with 0. 2ml highdose Sibiraea angustata extracts per every day;4)Medium dosegroup(3g/ml): the mice were transplanted tumor cell and perfused orallyinto stomach with 0.2ml medium dose Sibiraea angustata extracts perevery day;5)Low dose group(l. 5g/ml): the mice were transplanted tumorcell and perfused orally into stomach with 0.2ml low dose Sibiraeaangustata extracts per every day;6) CTX group: the mice were transplantedtumor cell and injected into abdominal cavity with CTX, 20mg/kg per every day. Expect normal control group, mice with tumor model were made with S180 ascites sarcoma. To observe mice survival quality, activity, psychosis, velvet, latent period of tumor growth, grow velocity and broken tumor during experiment. After fourteen days, the mice were killed by snapping neck. The tumor tissue were extracted carefully and weighed it at once and calculate the inhibition ratio to tumor . Then the tumor tissue were fixed by 10% formalin and made up paraffin section .To observe it in light microscopy. The level of expression of P53 -> Bcl-2 in tumor tissue werw measured by SABC Immunocytochemistry. To extract the spleen from the mice that were extracted the tumor tissue on germfree condition. The spleen were made one-celled mixture liquid. The level of IL—2 and TNF- a that was secreted by spleen cell were measured by ELISA.Result: extracts of medium and high dose can obviously suppress tumor growth and the inhibition ratio to tumor is 44. 6% and 40. 0%. Latent period of tumor growth is longer in every Sibiraea angustata group than model control group. Latent period of tumor growth is about 5 days.The scarfskin of broken tumor is littler hemorrhage than model control group. The mice psychosis and velvet are better than model control group. Some and many karyodieresis can be seen in light microscopy in model control group and low dose group and tumor cell grow actively. In high dose, medium dose group and CTX group, many tumor cell necrotized and presented pyknosis. Sibiraea angustataextracts of medium and high dose can promote the secretion of IL—2 and TNF-a . Amounts of IL-2 and TNF-a is 76. 100+4. 557. 80. 400 + 4. 719 pg/ml and 41.447 + 16. 373> 44.302 + 13.724 pg/ml. Compared with a blank model group, the mean diffence is significant at the 0.05 leve. Sibiraea angustata extracts of medium and high dose can suppress the express of mtP53 and the positive ratio is 54% and 52%;Sibiraea angustata extracts of high dose can suppress the express of Bcl-2 and the positive ratio is 82%. Compared with a blank model group, the diffence is significant at the 0.05 leve .Conclusion: 1. The Sibiraea angustata extracts can obviously suppress transplanted tumor-S180 growth in body.2. The mechanism of effect is associated with three aspects: 1) )The Sibiraea angustata extracts can bring forth tumor tissue necrose to some extent. 2) It can promote spleen cell secrete IL-2 and TNF-a in mice with tumor and enhance immune regulation to tumor. 3) It can suppress the expression of mtP53. Bcl-2 protein in tumor tissue and induce apoptosis of tumor cell.
