| The axillary buds of different breeds of Paeonia Suffruticosa were tested as explants in order to probe into the influence of different sterilized modes and time on explants.Different basic medias,regulators,accessories and environmental factors were studied in the course of inducement,multiplication and germination of axillary buds and the main influence factors and effective concentration scale were analysed.Healthy sterile test-tube plantlets were investigated in order to get the main influence factors and effective concentration scale in the inducement and multiplication course of embryonal callus.Anther was used as explant in order to that the main influence factors and effective concentration scale can be gotten in the inducement course.1, 0.1 HgCl2 was used as disinfectant and different sterilized modes and time were tested on explants.The results show:axillary buds is suitable to sterilized twice mode and time is better with 6min/6min. The stems with axillary buds is better with 6min. 2, The axillary buds of five breeds(P.suffruticosaWuLongPengSheng',P.suffruticosa 'LuoYangHong' ,P.suffruticosa' LuHeHong',P.suffruticosa' RouFuRong' ,P.suffruticosa' Zh ao Fen') were studied in the course of inducement culture.The results indicate:Significance difference is existed in different collecting time and in the five breeds.The collecting time is better with February and better effect of the five breeds are P.suffruticosa'WuLongPengSheng' and P.suffruticosa'LuHeHong'. 3 , Different basic medias, cytokinins and subculture times were studied in the course of multiplication of test-tube plantlets of two breeds (P.suffruticosa'WuLongPengSheng'and P.suffruticosa 'LuoYangHong').The results demonstrate:The suitable basic medias, cytokinins and subculture times are modified MS, TDZ and 3 respectively.And the suitable concentration of TDZ is O.lmg·L-1.Different lights and light intensity were studied in the course of multiplication and growth of test-tube plantlets of P.suffruticosaWuLongPengSheng'.The results show:LED light of which the proportion of light quality is adjustive had no effect on multiplication and growth.When the light intensity of general fluorescent was 30001x,it can promote the growth of test-tube plantlets effectively.CO2 fertilization and different concentrations of sucrose were tested in the course of multiplication and growth of test-tube plantlets of P.suffruticosa 'WuLongPengSheng'. |
