Effects Of Different Acupoints Of Wheat Moxibustion On Colonic Tissue Inflammatory Mediators And Mucosal Barrier In UC Rats

Objective:Establish the rats model of UC,treat with wheat-grain size cone moxibustion, to observe the rats from the angle of general morphology and molecular biology, to explore the similarities and differences between different acupoints as well as the mechanism of treating the rats with wheat-grain size cone moxibustion, and provide the reasonable acupoint prescription of treating UC with wheat-grain size cone moxibustion to the clinical.Material and method:Divided the 52 rats into control group(8 rats) and model group(44 rats) randomly. After using TNBS to eatablish UC rat model,eliminating two suspected intestinal obstruction rats, taking another two rats to validate the UC model was succeed, the remaining 40 rats were randomly dividied into model group, single acupoint moxibustion group, double acupoints moxibustion group, three acupoints moxibustion group, western medicine group, each group has 8 rats; single acupoint moxibustion group choose Zhongwan acupoint, double acupoints moxibustion group choose Zhongwan acupoint and Guanyuan acupoint, three acupoints moxibustion group choose Zhongwan acupoint、Guanyuan acupoint and Zusanli acupoint; take 1mg moxa to make them into moxa cone(bottom 2.5-3mm, high 4-5mm); use direct moxibustion, each acupoint use 7 moxa cones, one time a day, totally treated 7 times, the rats of control group and model group were fixed at the same time. Use scale to observe the Disease activity index(DAI) of rats before and after treatment, observe the intestinal tissue morphology change with light microscope, protein imprinting method to detect the expression of TNF-α and P38 MAPK, RT-PCR method to detect the expression of the tight junction protein Occludin、ZO – 1.Results:1.Before treatment, DAI statistical results show that : compared with the control group, the DAI of model group and each treatment group were significantly higher(P <0.05), there is no statistical difference(P>0.05) between the model group and each treatment group; After treatment, compared with the model group, the DAI of double acupoints moxibustion group, three acupoints moxibustion group and western medicine group were significantly lower(P <0.05 or 0.01); There are significant differences among the three groups of Wheat-grain size cone Moxibustion, compared the mean, the DAI of three acupoints moxibustion group was the lowest.2. Light microscope showed that the intestinal mucosal of control group was complete, the glands arranged neatly, with clear structure, no edema, congestion and tissue necrosis; while the intestinal mucosal of model group was missing, the structure of glands were irregular, submucosal edema, congestion and inflammatory cells infiltration;the glands of treatment group were arranged more neatly, the mucosa arranged slightly complete, the ulcer surface had new epithelial cells, and a small amount of inflammatory cell infiltration.3. The expression level of TNF-α and P38 MAPK result shows that: compared with the control group, the expression level of TNF-α and P38 MAPK of model group and each treatment group were significantly higher(P﹤0.01 or 0.05); compared with model group, the expression level of TNF-α and P38 MAPK of the three wheat-grain size cone moxibustion groups and western medicine group were significantly decreased(p < 0.01); there is statistically significant difference between the three treatment groups, compared the mean, the expression level of TNF-α and P38 MAPK of the three acupoints moxibustion group was the lowest.4. The expression level of Occludin and ZO-1 m RNA result shows that: compared with the control group, the expression level of Occludin and ZO-1 m RNA of the model group and each treatment group were significantly decreased(p < 0.01); compared with the model group, the expression level of Occludin and ZO-1 m RNA of the three wheat-grain size cone moxibustion groups and western medicine group were significantly higher(p < 0.01); there is statistically significant difference between the three treatment groups; compared the mean, the expression level of T Occludin and ZO-1 m RNA of the three acupoints moxibustion group was the highest.Conclusion 1. Wheat-grain size cone Moxibustion can reduce DAI of UC model rats、promote the repair of the intestinal mucosa, in our research the three acupoints moxibustion group and western medicine group have more obvious treatment effect.2. Wheat-grain size cone Moxibustion can decrease the expression of TNF-α and P38 MAPK of UC model rats, improve the expression of tight junction protein occludin、ZO-1, in our research the three acupoints moxibustion group and western medicine group have more obvious treatment effect.3. Wheat-grain size cone Moxibustion therapy may be through decreased the expression of TNF-α、P38MAPK and other cytokines, increased the expression of Occludin、ZO-1, thereby suppressing the intestinal inflammation, repairing the intestinal mucosal barrier function, achieving the purpose of treating UC.
