Wheat Grain Moxibustion Down-Regulates SREBP1c Through PI3k/Akt Signaling Pathway To Ameliorate Liver Lipid Metabolism In Hyperlipidemia Rats

OBJECTIVE:To explore the intrinsic mechanism of liver lipid metabolism in liver lipid levels in SD rats with hyperlipidemia model.METHODS:SD rats were used as subjects and fed with high fat diet for 8 weeks to induce the establishment of a hyperlipidemia model.ELISA,enzyme-linked immunosorbent assay kit was used to detect the serum levels of TG,TC,HDL and LDL in SD rats.After the establishment of the hyperlipidemia model,different interventions were carried out for each group.Times/day,2 days off every 5 days for 10 weeks.In the experimental group design,the blank group,the model group,the drug group and the inhibitor group were used to compare with the wheat grain moxibustion group.Intervention methods of each group:blank group was fed with basal diet,no other intervention;model group was fed with high fat diet without other intervention;drug group was fed with high fat diet,atorvastatin calcium was administered under isoflurane anesthesia;The moxibustion group was fed with high-fat diet,and under the anesthesia of isoflurane,the intervention of wheat grain moxibustion Zusanli;the inhibitor group was fed with high-fat diet,the intraperitoneal injection of LY294002 under isoflurane anesthesia and the intervention of wheat grain moxibustion Zusanli.After the 10-week dry end is expected,the material is taken and tested.1.Body weight and left hepatic lobe weight of SD rats in each group at 18 weeks after intervention.2.ELISA was used to detect the blood lipid components of SD rats,including serum TG,TC,HDL and LDL levels.3.Oil red O staining was used to detect lipid deposition in liver tissue.4.Immunohistochemistry was used to detect the distribution of SCAP and SREBP1c positive cells in liver tissue.5.Immunoblotting was used to detect the expression levels of PI3k,Akt,mTOR,CRTC2,SCAP,SREBP1c and other proteins in liver tissues.Real-time quantitative polymerization enzyme-linked reaction was used to detect the expression of SCAP mRNA and SREBP1c mRNA in liver tissue.RESULTS:The results were as follows:1 The wheat grain moxibustion Zusanli reduced the body weight and liver weight of hyperlipidemia SD rats;2 wheat grain moxibustion Zusanli reduced serum TG,serum TC and serum LDL in hyperlipidemia SD rats.At the same time,serum HDL was elevated;3 wheat grain moxibustion Zusanli improved the lipid deposition of liver tissue in hyperlipidemia SD rats;4 wheat grain moxibustion Zusanli increased the distribution rate of SREBP1 c positive cells in the liver of SD rats with hyperlipidemia However,the effect of SCAP positive cell distribution rate was not obvious;5 wheat grain moxibustion Zusanli activated PI3k/Akt signaling pathway in liver tissue of hyperlipidemia SD rats;down-regulated the expression of SREBP 1 c in liver tissue of hyperlipidemia SD rats,The effect of SCAP expression was not obvious;6 wheat grain moxibustion Zusanli up-regulated the relative expression of SCAP mRNA in the liver tissue of hyperlipidemia SD rats,and down-regulated the relative expression of SREBP1c mRNA.Conclusion:Wheat grain moxibustion Zusanli may down-regulate the expression of SREBP1c in liver tissue through PI3k/Akt signaling pathway,and reduce lipid metabolism in the liver,thereby improving liver lipid metabolism.
