Effect Of Moxibustion On Histomorphology And Serum ACTH And CORT In Kidney - Yang Deficiency Model Rats

Objectiv:Through moxibustion shenshu,guanyuan acupuncture point in rats model of kidney Yang deficiency, Observed the changes of kidney adrenal tissue morphological and the influence of serum ACTH, CORT to reveal the possible mechanism of moxibustion treatment kidney Yang deficiency.Methods:(1) Health40SD male rats were randomly divided into blank group, normal saline group, model group and moxibustion group,10rats in each group.(2) The blank group feeded conventional; normal saline group injected the physiological saline in hind leg muscle with0.3ml/d,28d continuous; Model group, the moxibustion group injected hydrocortisone about2.5mg/100g· d in hind leg muscle, made kidney Yang deficiency model for14consecutive days.Two groups continued to be hydrocortisone injection in order to maintain the kidney Yang deficiency syndrome, since15th day moxibustion group treated with moxibustion on shen shu, guan yuan point alternately, the other three groups rats also grab, bundling, for14consecutive days.(3) Take materials, Observe kidney and adrenal gland tissue morphology under optical microscope;RIA method to detect the leavel of serum ACTH and CORT.Results:1)After modelingl4days,the rats of model group and moxibustion rise less than control group significantly (P<0.05), model group and moxibustion group rectal temperature significantly lower than the control group (P<0.05), combined with the behavior of crouching, independent activities to reduce, frequent micturition, loose stool, scrotal moisture swollen, etc, which can illustrate the model to replicate the success.2)After14days of moxibustion treatment,compared with model group, moxibustion group average rat body quality growth and rectal temperature was significantly increased (P<0.05), but not as good as blank group significantly (P<0.05).3)It was shown that the morphological structure of adrenal gland and kidney of rats in model group had obvious glomerular damage, sparse distribution, renal tubular cell wall thinning, renal distal tubule expansion, proximal tubule atrophy of the adrenal cortex; irregular arrangement, unclear structure, thin cortex, medulla slightly thickened. By contrast to the model group,the morphological structure of kidney and adrenal gland in the moxibustion group, there was a certain degree of reduction.4)Adrenal quality and index:Model groups compared with the blank group and normal saline group comparison decreased significantly (P<0.01), Compared with model group, moxibustion group was increased with a high statistical significance (P <0.01), but moxibustion group compared with blank group was decreased significantly (P<0.01).5)Kidney quality and index:Compared with the blank group and the normal saline group,model group was decreased significantly (P<0.01); Compared with the model group,the quality of moxibustion kidney increased significantly (P<0.01), kidney index was increased,had a significance (P<0.05),But compared with the blank group, moxibustion group was decreased significantly (P<0.01).6)Serum ACTH results showed:Model group rats ACTH values compared with blank group, normal saline group were decreased significantly (P<0.01); moxibustion group compared with model group was increased significant (P<0.01), but lower than the blank group significantly (P<0.01),the blank group compared with normal saline group ACTH level had no difference (P>0.05).7)Serum CORT results showed:Serum CORT of model group reduced significantly compared with the blank group and normal saline group (P<0.01);Moxibustion group compared with the model group was increased significantly (P<0.01), but lower than the blank group significantly (P<0.01), the blank group compared with normal saline group CORT level had no difference (P>0.05).Conclusions:Moxibustion shen shu, guan yuan of acupuncture point can improve the kidney Yang deficiency, the symptom such as anus temperature, slow growth.The possible mechanism is that moxibustion shen shu, guan yuan of acupuncture point can promote the pituitary to secrete ACTH that can reduce the damage of shape and structure of kidney and adrenal gland on kidney Yang deficiency model rat and increase the quality of kidney and adrenal gland and index, and this may be one of the mechanism of action of moxibustion improve the symptoms of kidney Yang deficiency.
