We introduced Yacon to the northeast area of China, established method of seedling propagation through tissue culture technology. At the same time we discussed suitable transplantation period, the effect of film covering, feasible density and scientific fertilization. The results showed: MS+2, 4-D (2.0mg/L) +NAA (1.0 mg/L) was the best to inducing the callus of leaves; MS+BA (2.0mg/L) +NAA (0.20mg/L) was the best to inducing buds of stems; 1/2MS+NAA (1.0mg/L) +IBA (0.5mg/L) was the best to inducing the roots of regeneration plants. It was the most feasible for the growth of seedling of tissue culture when the ratio of glebe soil to sand to mold is 1:1:1. The survival rate reached 80%. The different transplant period made different effect on yield. The yield of earlier transplantation was higher than the later. It had no effect on the content of soluble total sugar and soluble protein, but it had effect on the content of Vc. The content of Vc of April 25th was the highest. It was 2.9504%. The plant height, leaf number and branch number of covering film were higher than CK. But the yield, the content of soluble total sugar, soluble protein and Vc were not obviously different. The yield per plant was the highest when the density was 80cm X 50cm. The content of total sugar was the highest. However the yiled per ha was the highest when the density was 80cm X 40cm. The content of protein was the highest. With the density increasing or decreasing, the yield was both decreased. The crucial factor to yield was content of N. The yield was the highest when the codes were 1.682, 1.000 and 1.000.. The yield was 20698.28kg/hm2. That was to say the level of N P K was fertilized by 240 kg/hm2 120 kg/hm2 , 240 kg/hm2. We selected the best scheme of fertilization through simulation of computer. We shall get the highest yield when the level of N, P K was fertilized by 202.47-221.09 kg/hm2 96.6-143.4 kg/hm2 193.91-286.09 kg/hm2 .The content of total sugar of exterior was about 2 times to interior. The content of total sugar was increased during storage. It increased about 3% during 15 days.
