| In this paper, the breeding process of a new combination of (03-10-6) pepper was expounded, the seedling technology of embryoid induction by anther culture of pepper, and four techniques of breeding and standardized cultivation were summarized.1 Breeding of a new combination of (03-10-6) pepperThe new combination of (03-10-6)pepper characteristics of late-maturing breeding for summer was hybridized by 00012 as the male and 02007 as the female. The time from planting to harvesting is about 60 days, the time from flowering to first harvesting is about 30 days. It has the characteristics of strong growth,80cm height,76cm plant expansion, strong branching ability, and strong ability of continuous fruit. Fruit has the characteristics of thick horn shape,20cm length,3-4cm diameter of fruit skin, and 50g average weight. It has dark green, and smooth peel. Its quality is good for soft texture and moderate peppery taste. Its yield, stable yield are good and its total yield is 4500 kg per 667 square meter.It is suitable for summer cultivation for its high resistance to virus disease, and resistance to anthracnose and blight at the period of its growth after many years of field observations.2 Initial establishment of the system of seedling technology of embryoid induction by anther culture of pepperThe suitable medium and culture conditions at the various stages of anther culture were summarized in this study. The anther of pepper was stripped and inoculated on induction medium of Q1 (MS+2,4-D 0.05 mg/L+KT 0.05 mg/L+sucrose 2.5%), and cultivated in dark 8 days at 34 ℃ in constant temperature incubator. And then the anther was transferred to Q2 culture medium (MS+KT 0.04 mg/L+2.5% sucrose) and cultivated at 25 ℃ in constant temperature incubator with the daily supplement of 10h light. After 20d inoculation, the white and young embryoid bodies were differentiated from the anthers. The embryoids were cut and cultivated in the medium until the height of seedlings are about 4-5cm. The seedlings were transplanted to the matrix of peat and pearl (2:1), and diploid plants of female parent 02007 of new combination of 03-10-6 were obtained by doubling. The seedling technology of embryoid induction by anther culture of pepper was established.3 Initial establishment of the system of tissue culture technology of cotyledon explants of pepperThe suitable medium,culture time,and culture conditions at various stages of tissue culture of cotyledon explants of pepper were summarized in this study.The cotyledon of pepper was inoculated on induction medium(MB5+AgNO3 4mg/L+PJ 5.0mL/L+6-BA 4.5mg/L+IAA1.0mg/L), then let the bud in the elongation medium(3.0 ng/LMB5+ 6-BA+IAA 1.5 mg/L+GA3 2.0 mg/L),and then placed on the rooting medium(1/2MS+ NAA 0.15mg/L+IAA 0.20mg/L).The system of tissue culture of cotyledon explants of pepper was established.4 Establishment of GAP cultivation techniques of new combination of (03-10-6)pepperThe suitable planting density,fertilization,and cultivation pattern of new combination of 03-10-6 pepper were studied.The GAP cultivation techniques were summarized for promotion of new combination of 03-10-6 pepper. |
