| Objective:In order to explore the EB virus and the correlation of the xinjiang region of Hodgkin’s lymphoma,understand the clinical status of patients with Hodgkin’s lymphoma in xinjiang region,based on the screening of Hodgkin’s lymphoma-related HLA-DPB1subtype in Xinjiang population,the binding mode,size and difference of HLA-DPB1subtype and Hodgkin’s lymphoma subtype and Epstein Barr virus gp42 in healthy population in Xinjiang region were further detected.Methods:1)We infected T1 cells with HLA-DPB1 subtype lentivirus by means of lentivirus infection,and constructed the recombinant vector of HLA-DPB1;2)gp42 protein was labeled by immunofluorescence labeling,and the binding efficiency of gp42 protein and HLA-DPB1 recombinant vector was explored at different concentrations to obtain the optimal binding concentration of gp42 protein;3)Flow cytometry was used to detect HLA-DP~+gp42~+double positive cells,and the affinity between HLA-DPB1 subtype and gp42 protein was calculated;4)We used molecular dynamics simulation to simulate the binding ofgp42protein and HLA-DPB1 subtype,and obtained the binding mode,binding form and basic properties of the complex formed by the two;5)were retrospectively analyzed in January 1,2016 to September 1,2020 for the first time in our hospital,105cases of patients with Hodgkin’s lymphoma in xinjiang,collect the patients clinical characteristics,laboratory results and pathological histology detection,using SPSS 16.0software for statistical analysis,to explore the EB virus infected with the EB virus infection between the clinical features and pathological characteristics;6)A total of 15patients with Epstein Barr virus-positive Hodgkin’s lymphoma and 10 patients with reactive lymph node hyperplasia were selected respectively to detect the expression of CD3,CD4,CD8,PD1,PDL-1,HLA-DPB1,LMP1,CD30 and other molecules in the two groups,and compare the differences in immune function and expression products.Results:1)The optimal conditions for lentiviral infection of T1 cells were selected as B2infective agent,MOI=100,and infection time of 72 hours.On this basis,plasmid vectors of LV-DPB1-H group and LV-DPB1-HL group were successfully constructed;2)The optimum binding concentration of gp42 protein was found,that is,the binding efficiency of HLA-DPB1 to gp42 was the highest when the concentration of gp42 protein was 80μg;3)The B chain of HLA-DPB1 binds to the A chain of gp42 through hydrogen bond to form a complex,which belongs to hydrophilic protein and has the Resolution of 3.25;4)The double positive rate of HLA-DP~+gp42~+in LV-DPB1-HL group was significantly higher than that in LV-DPB1-H group(29.467±2.108vs 15.867±0.929),and the difference was statistically significant;5)Clinical data and pathological characteristics of 105patients with Hodgkin’s lymphoma were collected and statistically analyzed.Univariate analysis showed that gender(P=0.007<0.1),age(P=0.000<0.1),Ann Arbor staging(P=0.088<0.1),and pathological type(P=0.000<0.1)were factors affecting the positive prognosis of Epstein Barr virus.Multivariate Cox regression analysis showed that age was an independent factor affecting the prognosis of EBV-positive Hodgkin’s lymphoma(P=0.000<0.05,HR:1.026,95%CI:1.014-1.042);6)Through the 15 cases of EB virus positive patients with Hodgkin’s lymphoma and 10 cases of patients with lymph node reactive hyperplasia of immune function and expression of product analysis,EB virus positive patients with Hodgkin’s lymphoma in the expression of LMP1,CD30 and PD-L1mass were higher than that of EB virus in patients with positive lymph node reactive hyperplasia,prompt LMP1,CD30,PD-L1 and the pathogenesis of EB virus positive patients with Hodgkin’s lymphoma correlation;Compared with the control group,the expression ratio of CD4~+T cells in the case group was increased,and the difference was statistically significant.The expression ratio of CD8~+T cells in the case group was decreased,and the difference was statistically significant.The expression level of HLA-DPB1 decreased in the proportion of case group vs control group,and the difference was statistically significant.Conclusion:EB virus gp42 protein can and human leukocyte antigen(HLA DPB1)on the cell surface,the combination of the gp42 protein concentration reaches 80μg,the combination of the two proteins can achieve higher efficiency,mutual combination can function within the cell,cause the body’s immune inhibition change,in the process of participation to the onset of Hodgkin’s lymphoma.In EB virus positive Hodgkin’s lymphoma group,gender,nationality,place,EBV DNA replication,Ann Arbor in installment,histopathological classification,the initial treatment and has no impact on the prognosis of patients with EB virus infection,but age,blood EB virus detection is the independent prognostic factors affecting the prognosis of patients with EB virus infection,and Epstein-Barr virus infection,the immune function of Hodgkin’s lymphoma and immune product lymph node reactive hyperplasia were changing.Therefore,we may be able to reduce the incidence of Hodgkin’s lymphoma by reducing the concentration of gp42,i.e.reducing the infection rate of Epstein-Barr virus.On the other hand,according to the results of molecular dynamics simulation,we can prevent the occurrence of the disease by breaking hydrogen bonds and preventing hydrophilic binding,etc.,and these data may provide some reference value for the treatment and vaccine research of Hodgkin’s lymphoma in the later stage. |
