Objective:The study is Baizhu Fuling Tang in different proportion in intervening experimental Crohn’s Disease rats with spleen-qi deficiency syndrome, through observing the effect on expression of cytokine, VIP and its recepter, TLR4and NF-κ B mRNA, to explore the immunologic mechanism and the optim proportion of Baizhu Fuling Tang on Crohn’s Disease.Methods:To establish the rat model of Crohn’s Disease with spleen-qi deficiency syndrome and cure model rats by Baizhu Fuling Tang in different proportion, TNF-a, IL-1β, IL-4, IL-10in rats blood serum, pathological changes and activity of VIP,VIPR1,TLR4, NF-κ Bofcolons were treated as observation and measurement indicators. And we detected those cytokinewith ELISA, protein activity of VIP and VIPR1byimmunohistochemistry,TLR4, NF-κ B mRNA with RT-PCR.Results:①Colon Organization and its Pathological Changes:Under light microscope, we observed there were ulceration, neutrophilic granulocyte infiltration, congestion, erosion, epithelium deletion, lamina propria gland lessen, vesselhyperplasia of the rates colons of model group. The experimental result showed the modelgroup was significant difference with the negative control group (P<0.05). Compared with model group, besides B1,B2, B3, B4, B7,B8, B9and B1O group, the rest groups were all significantly descended,②the level of TNF-a and IL-1β:Comparing to the negative control, the level ofTNF-a and IL-1β of the model group ascended significantly(p<0.05); Comparing to the model group:the level of TNF-a in Baizhufuling Tang B5group, B6group and B7group descended significantly (p<0.05); the level of IL-1β in BaizhufulingTang B6group and B7group descended signifcantly (p<0.05).③the level of IL-4and IL-10:Comparing to the negative control, the level ofIL-4and IL-10of the model group decended significantly (p<0.05); Comparing to the model group:the level of IL-4in Baizhufuling Tang B6group and B7group ascended significantly (p<0.05); the level of IL-10in Baizhufuling Tang B6group ascended significantly (p<0.05);rotein activity of VIP and VIPR1: Comparing to the negative control,the protein activity of VIP and VIPR1of the model group ascended significantly(p<0.05); Comparing to the model group:the protein activity of VIP and VIPR1in Baizhufuling Tang B5, B6and B7group decended significantly (p<0.05).⑤the level of TLR4and NF-κ B mRNA:Comparing to the negative control, the level of TLR4mRNA and NF-κ BmRNA of the model group ascended significantly(p<0.05); Comparing to the model group, the level of TLR4mRNA and NF-κ BmRNA in Baizhufuling Tang B5group, and B6group descended significantly (p<0.05). Conclusion:Baizhu Fuling Tang can intervene in experimental CD with stimulating the immune organs to excrete the immune cells through inhibiting activity of VIP and its receptor, TLR4/NF-κ B, thereby inhibiting the inflammatory response. The effect of Baizhu Fuling Tang B6group is better than other groups.Baizhu Fuling Tang can intervene in experimental CD through adjusting cytokine levels. The mechanism of action include:①Baizhu Fuling Tangregulates the balance withinflammatory and anti-inflammatory cytokines through suppressing IL-1β move and TNF-a secret, and promoting IL-4and IL-10to secret;②Baizhu Fuling Tang can regduce thelevelsof VIP and its receptor, TLR4/NF-κ Bof the experimental CD rats, and then regulate and control cytokine network.
