Objective:Observe the effect of Baizhufuling Tang with high/middle/low dose of baizhu and fuling on inflammatory factors of Crohn’s disease(CD) model rats with spleen qi deficiency,to study the best dose and immune mechanisms of Baizhufuling Tang curing Crohn’s disease model rats with spleen qi deficiency.Methods:In this study,we used spleen qi deficiency molding method attributed to qi exhaustion and improper diet combined with Morris method to replicate Crohn’s disease rat model with spleen qi deficiency. Dose of rats in accordance with the relevant provisions of state administration of traditional Chinese medicine (TCM), People after conversion unit weight of the rat is directly with the dose of20,10,5times as high, middle and low concentration to dose, and for intervention treatment.①Observing pathological morphology changes of colon tissue by visual inspection and light microscope.and scoring according to colonic mucosa damage index (CMDI) and colon histopathologic grading standard.②ELISA was used to detect the serum content of IL-1, IL-4, IL-6, IL-8, IL-10, and TNF-α.Result:Compared with blank control group,CD model group rats had significant difference in CMDI(P<0.05),serum content of IL-1, IL-6, IL-8, and TNF-α increased significantly(P<0.05),serum content of IL-4and IL-10decreased significantly(P<0.05). Compared with CD model group:①high and middle dose groups had significant difference in CMDI(P<0.05);②high/middle/low dose groups could down-regulate the expression of proinflammatory factor IL-1, IL-6, IL-8and TNF-α and up-regulate the expression of anti-inflammatory factor IL-4and IL-10;groups of middle dose had obvious effect of down-regulating the expression of IL-1, IL-6, IL-8and TNF-α(P<0.05);groups of high dose had obvious effect of down-regulating the expression of IL-1, IL-8and TNF-α(P<0.05);group of high and middle dose had obvious effect of up-regulating the expression of IL-4(P<0.05);group of middle dose had obvious effect of down-regulating the expression of IL-10(P<0.05). Conclusion:①It was successful to replicate CD rats model with spleen qi deficiency by spleen qi deficiency molding method attributed to qi exhaustion and improper diet combined with Morris method,estimating from molding factors,common manifestation,objective indicator,Counterevidence therapy and stability of model.②Baizhufuling Tang with high/middle/low dose of baizhu and fuling had certain regulating effect on inflammatory factors (IL-1, IL-4, IL-6, IL-8, IL-10and TNF-α)of Crohn’s disease (CD) model rats with spleen qi deficiency. By down-regulating the expression of IL-1, IL-6, IL-8, TNF-α and up-regulating the expression of IL-4, IL-10,and then relieving colon inflammatory reaction may be its therapeutic mechanism.③high/middle/low dose groups had certain regulating effect on inflammatory factors of Crohn’s disease (CD) model rats with spleen qi deficiency,but the group of middle dose had more comprehensive effect which may be related to its suitable dose.The results showed that there were different therapeutical effect of Baizhufuling Tang with high/middle/low dose of baizhu and fuling on Crohn’s disease (CD) model rats with spleen qi deficiency.Dosage dose of baizhu and fuling have certain dose-effect relationship with curative effect.This study provided experiment basis for clinic treatment of Baizhufuling Tang on CD.
