Effect Of Baizhufuling Tang With Different Proportions Of Baizhu And Fuling On Neuro-immune Mediator SP And Inflammatory Factors Of Crohn’s Disease Model Rat With Spleen Qi Deficiency

Objective:Observe the effect of Baizhufuling Tang with different propor-tions of baizhu and fuling on neuro-immune mediator SP and inflammatory factors of Crohn’s disease(CD) model rats with spleen qi deficiency,to study the neuro-immune mechanisms of Baizhufuling Tang curing Crohn’s disease model rats with spleen qi deficiency.Methods: In this study,we used spleen qi deficiency molding method attributed to qi exhaustion and improper diet combined with Morris method to replicate Crohn’s disease rat model with spleen qi deficiency. According to the maximum dose of baizhu and fuling recorded in Chinese Pharmacopoeia2010,we applied increasing-decreasing design method with baseline geometric proportion to regulate dose proportion of baizhu and fuling.we got ten different proportions of baizhu and fuling includinggroupsB1(0:27),B2(3:24),B3(6.21),B4(9:18),B5(12:15),B6(15:12),B7(18:9), B8(21:6),B9(24:3) and B10(27:0).The Crohn’s disease model rats with spleen qi deficiency were treated with ten different proportions of baizhu and fuling.②Observing pathological morphology changes of colon tissue by visual inspection and light microscope,and scoring according to colonic mucosa damage index (CMDI) and colon histopathologic grading standard.②ELISA was used to detect the serum content of IL-6,IL-8,IL-4and IL-10.③IHC was used to detect the content of SP and NK-1R in colon tissue.Result:Compared with blank control group,CD model group rats had significant difference in CMDI(P<0.05), serum content of IL-6and IL-8increased significantly(P<0.05),serum content of IL-4and IL-10decreased significantly(P<0.05),the content of SP and NK-1R in colon tissue increased significantly(P<0.05).Compared with CD model group:①different proportions groups had significant difference in CMDI(P<0.05);②all the different proportions groups could down-regulate the expression of proinflammatory factor IL-6and IL-8and up-regulate the expression of anti-inflammatory factor IL-4and IL-10;groups of B5,B6,B7had obvious effect of down-regulating the expression of IL-6(P<0.05);groups of B6,B7had obvious effect of down-regulating the expression of IL-8(P<0.05);group B6had obvious effect of up-regulating the expression of IL-4 and IL-10(P<0.05);group B7had obvious effect of down-regulating the expression of IL-4(P<0.05);③All the different proportions groups could down-regulate the expression of SP and NK-1RR in colon tissue,but groups of B5,B6,B7had obvious effect of down-regulating the expression of SP and NK-1R(P<0.05).Conclusion:①It was successful to replicate CD rats model with spleen qi deficiency by spleen qi deficiency molding method attributed to qi exhaustion and improper diet combined with Morris method,estimating from molding factors,common manifestation,objective indicator,Counterevidence therapy and stability of model.②Baizhufuling Tang with different proportions of baizhu and fuling had certain regulating effect on neuro-immune mediators (SP,NK-1RR) and inflammatory factors (IL-6,IL-8, IL-4, IL-1)of Crohn’s disease (CD) model rats with spleen qi deficiency.By down-regulating the expression of SP,NK-1R, IL-6, IL-8and up-regulating the expression of IL-4, IL-10,and then relieving colon inflammatory reaction may be its therapeutic mechanism.③Groups of B5,B6,B7had certain regulating effect on neuro-immune mediators and inflammatory factors of Crohn’s disease (CD) model rats with spleen qi deficiency,but the group B6had more comprehensive effect which may be related to its efficacy of tonifying spleen and eliminating dampness.The results. showed that there were different therapeutical effect of Baizhufuling Tang with different proportions of baizhu and fuling on Crohn’s disease (CD) model rats with spleen qi deficiency.Dosage proportions of baizhu and fuling have certain dose-effect relationship with curative effect.This study provided experiment basis for clinic treatment of Baizhufuling Tang on CD.
