Objective:To understand the effect of TongLuo TangTai Prescription on Diabetic Peripheral Neuropathy, evaluate it from the overall, molecular, and cell level while explain the effect of TongLuo TangTai Prescription treatment in JNK signal pathway of diabetic peripheral neuropathy in GK rats sciatic nerve tissue in terms of cell signal transduction, and finally open up new ideas and methods for the prevention and treatment of diabetic peripheral neuropathy medicineMethods:50 GK model rats were randomized to 5 groups:Control Group, high dose of TongLuo TangTai Prescription Group, middle dose of TongLuo TangTai Prescription Group, low dose of TongLuo TangTai Prescription Group, and Metformin and Mecobalamin Group.10 Wistar rats were used as nomal control group with ordinary feed, while the others were fed with high fat diet. The nomal and nomal control group were fed with normal saline 5. Oml/kg ·d, 1g; TongLuo TangTai Prescription groups were fed with 2.88g/kg ·d,1.44g/kg ·d,0.72g/kg ·d of TangTai Prescription, lg. Metformin and Mecobalamin group were fed with 100 mg/kg ·d, and 0.2mg/kg ·d, metformin and mecobalamin.28 days later, determinate the conduction velocity of sciatic nerve of rats by BL420 biological function experiment system, the serum tumor necrosis factor (TNF)-a of rats by ELISA; determinate C reaction protein (CRP); the sciatic nerve JNK,’and protein expression of caspase-3 with Immunohistochemical method; and detect sciatic nerve cell apoptosis by TUNEL. After 28 days, detect the fasting blood glucose, TNF-a, CRP, and sciatic nerve conduction velocity. Detected p-JNK, caspase-3 of rats by Immunohistochemical method. Detect the apoptosis:flow cytometry analysis.Results:Beginning of the experiment, The blood glucose was no statistical difference among GK rats, except comparing GK rats with Wistar rats(P<0.01). The blood glucose in each treatment group were decreased, high dose of TongLuo TangTai Prescription Group, middle dose of TongLuo TangTai Prescription Group, and Metformin and Mecobalamin Group were statistically different (P<0.05 or P<0.01) from the 2th week of th experiment. The blood glucose between treatment group and the Control Group were statistically different(P<0.01) from the 4th week of the experiment. End of the experiment, compared with the other groups, the blood glucos, TNF-α, CRP, sciatic pJNK, caspase-3 average IOD value, sciatic nerve apoptosis rate were all higher in Control Group (P<0.05 or P<0.01).So, TongLuo TangTai Prescription can reduce the blood sugar in diabetic peripheral neuropathy GK rats, decrese the level of TNF-α, CRP and inflammation, and inhibit the sciatic nerve expression of JNK and Caspase-3, and finally inhibit the apoptosis of neural cells, improve nerve conduction velocityConclusion:TongLuo TangTai Prescription can reduce The amount of drinking water and eating food of the rats, increase the weight, improve glucose metabolism, improve nerve conduction velocity, and has the Effect of anti T2DM inflammation and prevention of diabetic peripheral neuropathy. The effect may be achieved by reducing TNF-α and serum CRP concentration, inhibiting of JNK sciatic nerve and the expression of Caspase-3, reducing the apoptosis of nerve cells, thereby delaying the occurrence and development of diabetic peripheral neuropathy.
