Objective:By observing the effects of different concentrations of Tongluo Tangtai,which with the efficacy of promoting blood circulation and dissipating phlegm and dredging the collaterals,on inflammatory cytokines and nerve conduction velocity of GK mice with DPN, to find the best dose of Tongluo Tangtai to improve the inflammation and nerve function of mice with DPN. To provide data for Tongluo Tangtai used in the clinical treatment plan.Methods:After the 70 male GK rats,7-8 weeks old,were allowed to raised adaptively for 1 week,we selected 56 rats in this experiment with fasting blood glucose≥11.1mmol/L.And the selected rats would be given high fat diet for 12 weeks,so that they became a rat model of spontaneous DPN.According to different blood glucose levels,the 50 model rats were randomly divided into model group(n=10), Tongluo Tangtai high-dose group(n=10), Tongluo Tangtai medial-dose group(n=10), Tongluo Tangtai low dose group(n=10) and medicine (metformin +methycobal) group(n=10),a separate group of 7-8-week-old male Wistar rats which blood sugar were in the normal range,a total of six groups. They were given a period of 28 days of the drug separately. we observed the rats’general conditions,water intake,food intake and monitored the change of fasting plasma glucose (FPG) during the experiment.After the intervention.we would measure CRP, TNF-α and IL-6 levels in serum,sciatic nerve conduction velocity,HE staining pathological changes of the sciatic nerve of the rats in different groups.Results:The serum inflammatory factors (CRP, IL-6, TNF-a) levels of model group were significantly increased (P<0.01), the sciatic nerve conduction velocity of which step down obviously(P<0.01),and HE staining of sciatic nerve showed typical neuropathy pathological changes.Each dose group of Tongluo Tangtai could improve the general physical state,lower fasting blood sugar to some extent,reduce serum inflammatory factors’(CRP, IL-6, TNF-a) levels,improve sciatic nerve conduction velocity,and improve sciatic nerve tissue pathological damage of DPN GK rats.Experimental results showed that the efficacy of high-dose group Tongluo Tangtai was better than the medial-dose group and low dose group.Conclusion:The experiment results verified the feasibility of establishing an animal model of DPN in GK rats. Tongluo Tangtai with the efficacy of promoting blood circulation and dissipating phlegm and dredging the collaterals could restore and improve the growth of DPN GK rat,lower blood sugar levels, reduce inflammation reaction, improve nerve structure and conduction. Its impact on all observed indicators showed some characteristics of dose-response and high dose exhibited more advantageous results. |
