Objective:In this study, used immunohistochemistry and Western Blot technology,to detected the expression degree of nNOS proteins that be considered the plasticity functionalmarkers in developing of central neurons in the visual cortex area17and the lateral geniculatebody in light and visual input that stimulate visual central neuronal activity, compared to thevisual signal transmission function and protein expression in normal development rats,monocularly deprived rats and acupuncture rats, tested and verified the promoting effect ofacupuncture on the cortex and the lateral geniculate nucleus plasticity, to provide new ideasfor the mechanism research clarifies the acupuncture treatment of amblyopia.Methods:Clean14day old Wistar rats60, were randomly divided into the following5groups, with12rats in each group, respectively is: blank group, model group,cuanzhu through Jingming early-term acupuncture group, cuanzhu through Jingmingmedium-term acupuncture group and cuanzhu through Jingming late-term acupuncturegroup.With0.22mm×13mm needles acupuncture treatment, acupuncture to monoculardeprivation early-term acupuncture group at8days after, acupuncture to monoculardeprivation medium-term acupuncture group at15days after,acupuncture to monoculardeprivation late-term acupuncture group at22days after,cuanzhu to jingming spine5mm,with reinforcing reducing technique, daily treatment1times, each time for10minutes,the treatment period of7days. After the end of treatment were detected for P-VEP, and thenNOS protein expression were detected in the visual cortex area17and the lateral geniculatebody by immunohistochemistry and Western Blot technology, compared to the visual signaltransmission function and protein expression in normal development rats, monocularlydeprived rats and acupuncture rats.Results:After acupuncture treatment, compared to the early-term acupuncture groupand the middle term acupuncture group, P-VEP peak time and amplitude value decreases,there was statistically significant difference (P<0.05), so in the early and middle-term visualdeprivation cuanzhu through Jingming acupuncture can reduce the damage of visual deprivation; compared to the normal group and the model group the expression of visualcortex area17nNOS was statistically significant difference (P <0.05); Compared with themodel group, there is significant difference between the early-term acupuncture group andthe medium-term acupuncture group of the fluorescence intensity (P <0.05), and the late-term acupuncture group was not statistically significant (P>0.05). So in the early and middleterm visual deprivation cuanzhu through jingming acupuncture can promote the expression ofnNOS in visual cortex area17.In the normal group and the model group, the expression ofnNOS of the lateral geniculate body was found significantly different (P <0.05), comparedwith the model group, there is significant difference between the early-term acupuncturegroup and the medium-term acupuncture group (P <0.05), and the late-term acupuncturegroup was not statistically significant (P>0.05).The lateral geniculate nucleus in normalgroup, nNOS expression was significantly higher than that in the model group. Comparedwith the model group, acupuncture group, the early-term acupuncture group and themedium-term acupuncture group expression of nNOS was significantly higher than those inthe model group, the late-term acupuncture group nNOS expression was not significantlydifferent. So in the early and middle term visual deprivation cuanzhu through jingmingacupuncture can promote the expression of nNOS in visual cortex area17and lateralgeniculate body.Conclusion:(1) Acupuncture was better than traditional general way that treatment ofamblyopia, its clinical efficacy should be affirmed;(2) Cuanzhu through Jingmingacupuncture can change abnormal P-VEP wave in monocularly deprived rats;(3) Cuanzhuthrough Jingming acupuncture on early and middle stage of monocularly deprived rats ofnNOS in the visual cortex area17and lateral geniculate cells have a regulatory role, whichmay cuanzhu through jingming acupuncture intervening sleep deprivation damage ofbiological mechanism;(4)Cuanzhu through Jingming acupuncture has effects on theexpression of early and middle period of monocular deprivation group rat visual cortex area17and the lateral geniculate nucleus of nNOS protein, which may also cuanzhu throughJingming acupuncture intervening sleep deprivation damage of biological mechanism;(5)Cuanzhu through Jingming acupuncture activation associated factor activity, protect the lost light stimulation of the visual nervous system damage, promote the development of visualsystems, so as to improve the function of the visual system.
