Micro total analysis system is an expansive field, which roots in micro electro mechanical system, electronics, biochemistry and surface chemistry. The micro-channels, micro-chambers, micro-heaters and temperature sensors on the chip are fabricated by using MEMS technology on the substrate of glass, silicon and polymer for sample analysis. One important step of μTAS is the enrichment and separation of an analyte from its surroundings. Biological samples and ingredients are various, so enrichment and separation on a chip is remaining challenge.In this dissertation, the theory and recent development on biological sample pretreatment chip system have been systemic introduced. Micro biological sample pretreatment chip based on immunoaffinity chromatography was proposed, including chip design, microfabrication, surface modification and antibody enrichment realization.The topics of this dissertation are presented below:1 Research and development on biological Sample pretreatment chip is reviewed. Then new biological sample pretreatment chip based on immunoaffinity chromatography and research projects were proposed.2. Biological sample pretreatment chip based on silicon was designed and fabricated. To increase the contact surface, intersectant micro-pillar array structure was produced in the channels of the chip. The contact surface increase up to 2 times than channels without micro-pillar array. Velocity distribution in intersectant micro-pillar structure was more uniform than in parallel micro-pillar structure.3.
