To explore the role of HLA-DRB1 and its upstream regulatory regions in the pathogenesis of rheumatoid arthritis(RA) and the correlations between HLA-DR alleles and the susceptibility, clinical manifestations of RA patients.2). The progression and prognostic markers of RA were analyzed clinically and with X-ray.Methods:1). 86 patients and 106 race matched controls were studied in which HLA-DR types were perfomed by the method of DNA amplification with sequence-specific primers (PCR-SSP).2). The subtypes of HLA-DR4 were determined by the method of hybridization of PCR products with sequence-specific oligonucletides (PCR-SSO).3). The polymorphism of DR4 gene upstream regulatory regions were determined by the methods of single-stranded conformational polymorphism (SSCP) and DNA sequencing.4).
